The Expression Of KRT2 Gene In Different Colour Alpaca Skins And Its Effect On Melanin Production

The expression profile of sheep skins with different hair colors showed the different mRNA expression of KRT2,which suggested KRT2 might be relative to the hair color formation.To investigate the effect of KRT2 on skin and the pigment production of alpaca.First using immunohistochemistry(IHC)experiment,we found that KRT2 was expressed in hair follicle root sheath and hair ball in alpaca skin by immunocytochemistry,and brown alpaca skins of KRT2 gene expression higher than white alpaca skins.real-time quantitative PCR(qRT-PCR)and Western blotting(WB)analysis revealed that mRNA and protein expression of KRT2 gene in brown skin was higher than that of the white alpaca skin,and the difference is significant(P<0.01).In addition,we cultivated alpaca melanocytes,transfection with KRT2 in alpaca melanocytes,through the construction of overexpression vector deal with alpaca melanocytes after 72h,Measure the microphthalmia-associtated transcription factor(Mitf),tyrosinase(Tyr)and tyrosinase-related protein 1(Tyrpl)genes mRNA and protein expression of KRT2,Compared with the control group,exposure to KRT2 with the overexpression led to up-regulation of both mRNA and protein levels of these three genes in alpaca skin melanocytes(P<0.01);Spectrophotometry measuring A490 proved that pmirGLO-KRT2 handling expression of alpaca skin melanocytes produce melanin content are respectively pmirGLO-NC cell group 1.45 times(P<0.01)and the blank cells group 1.36 times(P<0.01),while pmirGLO-NC cell group and the blank cells group no significant difference.All the datas suggest that the KRT2 may promote melanogenesis in alpaca skin melanocytes through stimulating melanin-formation signals in the alpaca skin melanocytes,which provided the necessary theoretical basis for further exploration of melanin production.
