Cucmber is one of the most important vegetables in China.The production and area harvested of cucumber in China are ranking first.Aphid is one of the main pest in the growth of cucumber,which seriously infect the growth,production,quality and benefit of cucumber by sucking the nutrients,spreading of the virus disease and other ways.Using chemical pesticides to control aphid is the main method in production,but large amount of chemical pestickdes had brought many problems,for example,enhance the resistance to chemical pesticides,pollution of agricultural ecological environment,killing aphid enemies and pollination insects and so on.Using iTRAQ technology(Isobaric Tags for Relative and Absolute Quantitation),aphid resistance proteins in cucumber were screened and identified between aphid treatment(5 apterous adult melon aphid)and no-aphid treatment at the 6th day after aphid infection.Treated by jasmonic acid,the difference between 80 apterous adult melon aphid treatment and Control and expression of related genes were studied in this experiment.The main research results are as follows:Fifty-four differential expression proteins were screened out by iTRAQ.Among these,19 proteins were up-regulated,and 35 proteins were down-regulated.Nine of 54 were chosen to do Quantitative Real-Time Polymerase Chain Reaction,(qRT-PCR),which showed the trend of gene expression and protein expression were basic unanimous.DEPs were gathered to metabolic process,cellular process,single-organism process and etc.in biological process with GO(Gene Ontology)analysis.In cell component,DEPs were gathered to cell,organelle,macromolecular and etc.In molecule function,DEPs were gathered to binding,catalytic activity and etc.With KEGG(Kyoto Encyclopedia of Genes and Genomes)analysis,DEPs in cucumber after aphid infection were gathered to ribosome,starch and sucrose metabolism,flavonoid biosynthesis,glutathione metabolism,phenylpropanoid biosynthesis,alpha-linolenic acid metabolism,carbon metabolism and so on.Some DEPs had relationships with each other,for example,gene co-expression.Using jasmonic acid treating the cucumber,the best performance was shown in the 1 mmol/L JA treatment and the number of aphid in treated cucumber were significant lower than untreated cucumber,which shown the plant resistance ability after jasmonic acid treatment.The Csa2G395815,gene encoding up-regulated protein 12-oxophytodienoate reductase 11,was up-regulated both in treatments of different number of aphid infection and different jasmonic acid concentration.Besides,1-aminocyclopropane-1-carboxylate oxidase homolog 6-like,peroxidase 2,anthocyanidin 3-o-glucosyhransferase 2-like,scopoletin glucosyltransferase-like,pathogenesis-related family protein and pectinesterase-like,these 6 proteins were up-regulated both in protein expression and gene expression;Chalcone-flavonone isomerase family protein and Flavanone-3-Hydroxylase were down-regulated both in protein expression and gene expression.
