Improvement of meat quality and excessive deposition of abdominal fat are the 2 kinds of urgent problem in broiler nutrition regulation.C type natriuretic peptide(CNP)is one of the important member of the natriuretic peptide family,It plays an important role in the development of muscle and fat deposition of mammalian,but its specific molecular mechanism is rarely reported in chicken.The preadipocytes of abdominal fat and Intramuscular fat induced by CNP,By comparing and analyzing the changes of the proliferation,differentiation and decomposition ability of the two kinds of adipocytes,the molecular genetic mechanism of chicken lipid metabolism regulated by CNP was studied from cellular level.Experiment 1: The lipid metabolism of adipocytes of chicken abdominal fat regulated by CNP.In this experiment,the preadipocytes was extracted from abdominal fat tissue of chicken,and the preadipocytes induced by CNP.The proliferation,differentiation and deposition ability of preadipocytes determined by MTT,Edu and the red O staining.The expression of NPRA,NPRB,NPRC mRNA determined by Real-time fluorescent quantitative PCR.The cGMP concentration determined by ELASA.The results showed : 1)The preadipocytes induced 24 h and 48 h by CNP.The proliferation of preadipocytes was significantly higher than the control group(P < 0.05).2)The preadipocytes induced 3d and 6d by CNP.The differentiation of preadipocytes was significantly lower than the control group(P < 0.05),but the concentration of glycerol in the medium was significantly higher than the control group(P < 0.05).There was no significant change in the expression of NPRA mRNA,but the expression of NPRB and NPRC mRNA increased significantly(P < 0.05),and the concentration of cGMP in adipocytes was significantly higher than the control group(P < 0.05).These results suggested that the proliferation and differentiation of preadipocytes,and decomposition of adipocyte may regulated by CNP activated the NPRB/NPRC-cGMP signaling pathway.Experiment 2: The lipid metabolism of adipocytes of chicken intramuscular fat regulated by CNP.In this experiment,the preadipocytes was extracted from breast muscle tissue of chicken,and the preadipocytes induced by CNP.The proliferation,differentiation and deposition ability of preadipocytes determined by MTT,Edu and the red O staining.The expression of NPRA,NPRB,NPRC mRNA determined by Real-time fluorescent quantitative PCR.The cGMP concentration determined by ELASA.The results showed : 1)The preadipocytes induced 24 h and 48 h by CNP.The proliferation of preadipocytes was significantly higher than the control group(P < 0.05).2)The preadipocytes induced 3d and 6d by CNP.The differentiation of preadipocytes was significantly lower than the control group(P < 0.05),but the concentration of glycerol in the medium was significantly higher than the control group(P < 0.05).There was no significant change in the expression of NPRA mRNA,but the expression of NPRB and NPRC mRNA increased significantly(P < 0.05),and the concentration of cGMP in adipocytes was significantly higher than the control group(P < 0.05).These results suggested that the proliferation and differentiation of preadipocytes,and decomposition of adipocyte may regulated by CNP activated the NPRB/NPRC-cGMP signaling pathway.
