In this paper,the orthogonal extraction method was used to optimize the extraction process of rosmarinic acid in Meaona chinensis Benth.The bacterial anal swabs were collected from clinically occurring pet dogs and the drug resistance genotypes which were detected.The double microdilution method was used to determine Meaona chinensis Benth.The MIC of intestinal bacteria isolated in clinic and the use of trace checkerboard dilution method to determine the antibacterial effect of Meaona chinensis Benth water extract and antibacterial drugs on clinical isolates of intestinal bacteria,and use alkaline phosphatase(AKP),SDS PAGE,soluble The concentration of protein and Nucleotide leakage were used to preliminarily study the antibacterial mechanism of Meaona chinensis Benth extract against clinical isolates of intestinal bacteria.The main results of the test are as follows:1.In the optimized extraction process,single factor tests were firstly conducted,and orthogonal tables were designed using SPSS to conduct experiments.The final optimization process was:ethanol 40%,solid-liquid ratio 1:10,ultrasonic power time 45 min,and maximum extraction rate of 16.7%.2.The anal swabs were collected from naturally-occurring dogs at Animal Hospital of Guangxi University,and finally identified as Proteus,Klebsiella pneumoniae,and E coll.Among them,Proteus contains SHV type,E coli contains SHV type and fosA3 which is resistant to fosfomycin are both multi-drug resistant and ESBLs-producing strains.3?The MIC of Proteus spp,Klebsiella pneumoniae,and Escherichia coli was 7.8 mg/L,125 mg/L,and 500 mg/L,respectively,and they were synergistically used with these antibiotics for these three bacteria.The result of this experiment was additional and synergetic.4?The extract of Meaona chinensis Benth can inhibit or eliminate the synthesis of total bacterial proteins;the increase of AKP actibity and solublepro;teincontent in themedium and play a destructive role on the cell membrane and cell wall of E.coli.5?Four virulence genes were detected by PCR in E.coli,namely marA,csgA,sodA,and Issl.Mice in the model group had different degrees of infection,and the red blood cell and hemoglobin were statistically significant compared with the model group(P<0.05);Spleen hemorrhage can be seen from the section.Pathological changes such as inflammatory cells,congestion,and cell cavitation were observed in the liver of the liver model group.After treatment with the crude extract of Meaona chinensis Benth,the contrast model group was less congested,and the spleen and liver were present,which could be regarded as a certain degree of protection.
