Zizania latifolia which belonging to Zizania Gronov.ex L.of Gramineae family is a perennial grass.Zizania latifolia.It can be propagated through seeds but also by tillering reproduction.It is widely distributed in China and its traces of growth extend from the south to Hainan and north to the vast regions of Heilongjiang.The Zizania latifolia is a good garden landscaping and landscaping plant;its Chinese Wild Rice is rich in nutrients and used by people as a high-quality cooking ingredient;after the stalk is infected with smut,the tissue enlargement can be used as a vegetable by people;Its gene has high resistance and characteristics and is an exogenous excellent gene pool for rice breeding.Liangzi Lake has a special formation,unique topographic structure,and rich biodiversity.Since the records in 1955,there have been investigations on the Liangzi Lake plant community.At present,there are no detailed studies and investigations on the genetic diversity of wild salmon resources in Liangzi Lake.In this study,a comprehensive and detailed survey was conducted on wild Zizania latifolia resources in Liangzi Lake in order to improve the basic data of wild Zizania latifolia resources in Liangzi Lake region,and to clarify the situation and genetic diversity of wild Zizania latifolia resources in the region,and to provide a basis for the local wild Zizania latifolia resources.Protection and use provide a certain basis,the research results are as follows:This study based on the topography of Liangzi Lake,uses previous literature review,visits,and other forms in combination with field survey collection.A survey of wild Zizania latifolia resources in Liangzi Lake was conducted around the lake.Samples of 42 populations of wild Zizania latifolia from Liangzi Lake were collected from the field survey,and the study also has recorded the water quality,habitat,growth status,and plant size of the area in order to analysis and description in later study,This essay combined with the actual situation,put forward some views and conclusions for the future.19 pairs of paddy have been marked select out from the 1113 pairs paddy Simple Sequence Repeat(SSR)by Wu Guolin,and then selected 9 pairs of clear and stable Simple Sequence Repeat(SSR)to analyze the genetic diversity of wild Zizania latifolia resources in Liangzi Lake.108 test specimens were collected from 42 wild Zizania latifolia populations and 71 bands were amplified.Among them,70 bands were polymorphic bands and the polymorphic loci is 98.59%.Analysis with Pop gene showed that the genetic distance between populations ranged from 0.0540 to 0.8226,and the average genetic distance is 0.2674.Shannon information index I* is 0.506 between populations,and Nei's gene diversity index He is 0.336.Using UPGMA method in MEGA software to divided the 42 populations into 4 groups,which is consistent with main ingredient analysis of PCA results.Select 13 pairs of clear and stable inter-simple sequence repeat(ISSR)which was optimized and established by Wu Guolin were used to analyze the genetic diversity of wild Zizania latifolia in Liangzi Lake.108 test specimens were collected from 42 wild Zizania latifolia populations and 72 bands were amplified.Among them,62 bands were polymorphic bands and the polymorphic loci is 86.1%.Analysis with Pop gene showed that the genetic distance between populations ranged from 0.0626 to 0.6272,and the average genetic distance is 0.244.Shannon information index I* is 0.4449 between populations,and Nei's gene diversity index He is 0.2963.Using UPGMA method in MEGA software to divided the 42 populations into 4 groups,which is consistent with main ingredient analysis of PCA results.After the comparison of SSR and ISSR data,At the the detection of information volumethe,ratio of polymorphic sites of SSR was 12.49% higher than the ISSR.So SSR had higher analysis efficiency than ISSR.In the comparison of genetic distance and genetic similarity coefficient,the data showed that the results obtained by the SSR and ISSR were very similar.Both SSR and ISSR in the AMOVA molecular variation analysis showed that wild Zizania latifolia population variation in Liangzi Lake occurred within the population.In the cluster analysis results of the two method both dicided the 42 wild populations into 4 categories,and 33 of the 42 germplasm resources were clustered in the corresponding category,with a coincidence rate of78.57%.
