Objective:The optimum concentration of methyl jasmonate and the frequency of stimulation induced Aquilaria sinensis to form agarwood were probed to optimize the artificial agarwood-inducing technology and to obtain high quality agarwood in a short time.Methods:?1?After repetitive stimulation?only one time,1 time per month,2 times per month,4 times per month?of A sinensis with 2 kinds of concentrations of methyl jasmonate(10,50 g·L-1)for 6 months.Determine the content of agarotetrol in the eaglewood by high performance liquid chromatography.?2?After repetitive stimulation?2 times per month?of A.sinensis with 5 kinds of concentrations of methyl jasmonate(2,5,10,20,50 g·L-1)for 1-6 months.Determine the content of agarotetrol by high performance liquid chromatography,and the content of alcohol soluble extracts by the method of hot leaching under the term of alcohol soluble extractives,then evaluate the quality of the agarwood.?3?A.sinensis was induced with 5 g·L-1 methyl jasmonate by 2 times per month.And the macroscopical and micro xylem structures of the injured parts of 0,3,5,8,10,15 and 30d were observed.Determine the content of alcohol soluble extracts by the method of hot leaching under the term of alcohol soluble extractives.The mechanism of A.sinensis to form agarwood was preliminarily analyzed.Results:?1?The content of agarotetrol in the agarwood of A.sinensis treated by 2 times per month with 10 g·L-1 concentration of methyl jasmonate is the highest,1.78 mg·g-1.?2?The content of agarotetrol in the agarwood of A.sinensis treated by 2 times per month with 5 g L-1 concentration of methyl jasmonate for 6 months is highest,3.08 mg·g-1,but the content of alcohol soluble extracts treated by 2 times per month with 10 g·L-1 concentration of methyl jasmonate for 6 months is the highest,30.99%.?3?After the injury of the white wood,the color of the injured part became yellow and gradually brownish,and the content of the alcohol soluble extractive in it increased gradually,and the yellow grease could be observed in 15?30 days in optical microscope.?4?In 6 months,the content of agarotetrol and the content of alcohol soluble extract increased with the prolongation of continuous stimulation time,and the growth rate was quite stable.Conclusions:?1?Methyl jasmonate has a good effect on the induction of A.sinensis to form eaglewood,and the effect of repeated stimulation on A.sinensis was 2 times per month>1 times per month>4 times per month>only once.5 g-L-1 methyl was most beneficial to improve the content of agarotetrol,but 10 g-L-1 methyl jasmonate is most conducive to improve the content of alcohol soluble extract in the eaglewood.?2?After being subjected to stress,A.sinensis started defense reaction.The xylem secreted and accumulated defensive substances,and formed yellow oil gradually that could be observed within 15?30 days.
