Among many negative regulators of muscle growth,myostatin plays a significant role.Myogenin is an important member of the MyoD gene family.Although there have been some studies on myostatin and myogenin in mammals,many of its functions in fish are not clear.In order to study the model of Trachinotus blochii muscle growth,we clone the Trachinotus blochii MSTN-1,MSTN-2 and MyoG by RACE and transcriptome,which the gene about muscle growth.We compared the MSTN-1,MSTN-2 and MyoG phylogenetic relationships and homology.The transcripts of MSTN-1,MSTN-2 and MyoG genes were measured in different tissues.The expression level of MSTN-1,MSTN-2 and MyoG mRNA in midbrain and muscle in different temperature,and the expression level of MSTN-1,MSTN-2 and MyoG mRNA in midbrain and muscle during the starvation and refeeding.1.In the present study,full length cDNA sequences of MSTN-1,MSTN-2 and MyoG were cloned.The MSTN-1,MSTN-2 and MyoG cDNA contained the open reading frame of 1131,1080 and 753 bp,encoding the polypeptides of 376,359 and 250 amino acids,respectively.MSTN-1,MSTN-2 and MyoG were high similarity with Pampus argenteus by comparison of phylogenetic tree and homology.2.The transcripts of MSTN-1,MSTN-2 and MyoG genes were measured in different tissues.MSTN-1 transcript was mainly expressed in muscle,brain and fat,while MSTN-2 and MyoG transcripts were detectable mainly in brain and muscle,respectivtly.3.The expression levels of MSTN-1,MSTN-2 and MyoG genes in midbrain and muscle were determined under four temperatures(21,25,29 and 33?).The growth of the T.blochii and the expression of the three genes in the midbrain and muscle were studied under different feeding methods.In the temperature control experiment,MSTN-1 gene had the highest expression at 29?,MSTN-2 was 33?,MyoG was 25?.The fitness temperature of muscle was 25-29?.4.In the feeding mode,the MSTN gene was increased during starvation and decreased during refeeding.MyoG had a different expression model with other5.Fishes during starvation.After the hunger,there was a compensatory growth.Through the experiment,for the future production to find the most suitable breeding temperature and the optimal feeding mode provides experimental basis.
