Chlorella Growth Factor?CGF?attracts attention in many research fields such as food,medicine,health care and feed,because of its high nutritional value,antioxidant resistance,enhanced immune function,inhibit cancer cells,and many other physiological functions.In this study,Chlorella vulgaris was used as the research object.By optimizing its culture conditions?temperature,ratio of nitrogen and phosphorus,pH,light-dark cycles?,to obtain high yield of Chlorella vulgaris the best culture conditions.The main components of CGF are qualitative and quantitative analysis of monosaccharides,amino acids,fatty acids,vitamins,and proteins,and a preliminary analysis of protein function.Chlorophyll fluorescence technology was used to study the effects of temperature,nitrogen-phosphorus ratio,pH,light-dark cycles on the growth and photosynthesis of Chlorella vulgaris.The results showed that the experiment temperature,nitrogen and phosphorus ratio,pH and light dark cycle had significant influence on the growth of Chlorella vulgaris and the state of photosynthesis?P<0.05?.?1?Temperature of 15?,Chlorella vulgaris growth and chlorophyll fluorescence parameters have obvious inhibition,at 20?,25?and 30?,35?Chlorella vulgaris cell density and chlorophyll fluorescence parameters were increased with the extension of incubation time,the temperature of 30?Chlorella vulgaris growth and photosynthetic state is best.?2?In the treatment group of nitrogen and phosphorus,the values of Fv/Fm,Fv/F0,?PS?,Yield,ETR were all increasing in 08d,began to decline after 8d,in nitrogen and phosphorus ratio is 17.30:1 the best treatment group of Chlorella vulgaris growth and photosynthesis.?3?PH 9,chlorella vulgaris cell density and chlorophyll fluorescence parameters is restrained,the rest of the treatment group the indexes with the extension of incubation time steadily rising trend,pH is 7the best treatment group of Chlorella vulgaris growth and photosynthesis.?4?The light-dark cycles was 14hL:10hD,the growth and photosynthesis of chlorella vulgaris were the best,and the effect of cell density and chlorophyll fluorescence parameters was from large to small in order of 14hL:10hD,12hL:12hD,16hL:8hD,10hL:14hD,8hL:16hD.This study provides a data base for selecting the best environmental conditions for the growth of chlorella vulgaris.The content of monosaccharide,amino acid,fatty acid,vitamin and protein in CGF was determined by HPLC.The main results are as follows.?1?The contents of CGF monosaccharide contained glucose,galactose,mannose,rhamnose,arabinose and fucose,etc.,with the content of 3.601 mg/kg,2.495 mg/kg,0.457 mg/kg,0.831mg/kg,0.833 mg/kg and 0.429 mg/kg.?2?CGF contains 35 kinds of fatty acids,including decanoic acid,undecanoic acid and lauric acid,which are respectively4.33mg/kg,0.72mg/kg,10.84mg/kg,etc.?3?CGF contains 17 kinds of amino acids including aspartic acid,glutamate and cysteine,the content is 30.83g/kg,25.47g/kg,2.51g/kg,etc.?4?The contents of VA,VB1,VB3,VB6,VC and VD22 in vitamins were85.56mg/kg,156.39mg/kg,2690.25mg/kg,1508.22mg/kg,2972.17 and 22.28mg/kg.?5?CGF contains 720 kinds of protein,the highest amount of protein is the protein CHLNCDRAFT32555[Chlorella variabilis].The results of this study preliminarily analyzed the main components of CGF and laid a foundation for further study on CGF function.Using OmicsBean omics data integration analysis cloud platform,the protein group analysis of the screened CGF protein was performed.?1?GO function analysisfound that the enrichment of chlorella protein CGF in 397 biological process,123 cell component and 227 molecular functions,including significant enrichment process of biology,cell composition,molecular functions respectively 165,62 and 98.?2?KEGG Pathway enrichment analysis showed that CGF protein was enriched to Carbon fixation in photosynthetic organisms and other 55 metabolic pathways,the significant metabolic pathway was 16.?3?The protein interaction involved the 143edges of 50 nodes,the highest protein connection pathway is Metabolic pathways,and47 genes were involved in Metabolic pathways,and the protein with the highest degree of network connectivity was the protein CHLNCDRAFT33230[Chlorella variabilis].This study lays a theoretical foundation for further research on the regulation mechanism of CGF.
