Abstract/Summary:
Objective:To study the effect Bacillus coagulans on Gut Microbiota in male Kunming Mice.At the same time,to explore the changes of Gut Microbiota after the E.coli infection.Method:The control groups were administered with normal saline(0.9%w/v sodium chloride).Low-dose group,middle-dose group and high-dose group were administered with 106 CFU,107CFU,108 CFU of Bacillus coagulans respectively.After 30-d administration,We collected fecal samples of cecum in order to analyze the gut microbial community by high-throughput sequencing.After Intraperitoneal injection of Eoli on the treated mice(CE,TEL,TEM,TEH group)and 0.9%w/v sodium chloride(CG1 group),We Analysis the differences between the 5 groups,analysis of five groups of cecum content difference of intestinal flora in mice.Result:The results showed that:1)To some degree,the high dose group can increase the microbial diversity and richness but it can reduce the complexity of Gut Microbiota;2)The PCoA,PCA and NMDS analysisindicated that significant difference were observed after The Bacillus coagulans was fed.However,the feeding concentration had little effect on it.3)The Bacillus coagulans of low,middle and high dose can significantly increase the richness of Proteobacteria(p<0.05).The Paracocccus make a difference.The high dose group can significantly increase the richness of Verrucomicrobiaceae(p<0.01)and Ruminiclostridium9.The middle dose group can significantly reduce the richness of Bacteroides,Prevos,and Actinomycetes(p<0.05).4)The deaths accounted for 30%in group CE,however,there is no deaths in the rest of the group.5)After Intraperitoneal injection of Eoli,there were significant falling in complexity of Gut Microbiota,The Deltaproteobacteria(p<0.01)and the Rhodospirillaceae(p<0.05)are significantly falling.6)The feeding of Bacillus coagulans before Intraperitoneal injection of Eoli can significantly increase the relative content of Alcaligenaceae and RikenellaceaeRC9(P<0.05),it can significantly falling the relative content of Bacteroidetes and Porphyromonadaceae and Rikenella than the group of no Eoli.7)The feeding of Bacillus coagulans before Intraperitoneal injection of Eoli can significantly increase the relative content of Firmicutes and Clostridia and Clostridiales and Lachnospiraceae and Alcaligenaceae than the group of no Bacillus coagulans group;it can significantly falling the relative content of Proteobacteria and Gammaproteobacteria and Bacteroides and Parabacteroides.Conclusion:The test of Gut Microbiota by using Illumina Hiseq high-throughput sequencing after feeding Bacillus coagulans a period of time,the Bacillus coagulans can adjust Gut Microbiota under the premise of without changing advantage bacterium group.The Intraperitoneal injection of Eoli,can significantly increase the relative content of Firmicutes and increase the intestinal flora diversity and maintain a healthy intestinal tract environment,so the Bacillus coagulans has protective effect under the attack of Escherichia-Shigella.
