Duck viral hepatitis(DVH)is an acute,highly lethal infectious disease.In recent years,pancreatitis-type duck viral hepatitis,which is characterized by yellow pancreas,and cause greate conomicloss to the duck in dustryObjective:In the experiment,180 Cherry Valley ducklings were infected with hepatitis-type duck hepatitis A virus(DHAV)and pancreatitis-type DHAV respectively,the apoptosis of hepatocytes and pancreatic epithelial cells,changes of hepatic function and pancreatic function were compared.The purpose is to investigate the differences of lesions between pancreatitis-type duck viral hepatitis and hepatitis-type duck viral hepatitis,and the pathogenesis of pancreatitis-type duck viral hepatitis.It can also provide evidence for the differential diagnosis of pancreatitis-type duck viral hepatitis.Methods:7-day-old ducklings were infected with hepatitis-type DHAV(FZ86 strain)and pancreatitis-type DHAV(ZJ1206 strain)respectively,blood samples were collected respectively at 12,24,36,48,120,168 and 216h post-infection,liver and pancreas were collected respectively at 24,72,120 and 168h post-infection.The apoptosis of hepatocytes and pancreatic epithelial cells were detected by situ end labeling technique(TUNEL),the concentration of caspase-3 in liver and pancreas were detected by enzyme-linked immunoabsorbent assay(ELISA)method,and alanine transaminase(ALT),aspartate aminotransferase(AST),pancreatic amylase(AMYL)and lipase(LIPA)were detected used automatic biochemical analyzer.Results:The number of apoptotic hepatocytes in ducklings infected with hepatitis-type DHAV were more than that of infection with pancreatitis-type DHAV,the difference was significant(p<0.05).The most apoptosis of hepatocytes in ducklings infected with hepatitis-type DHAV occurred at 24h post-infection.While the number of apoptotic pancreatic epithelial cells in ducklings infected with pancreatitis-type DHAV were more than that of infection with hepatitis-type DHAV,the difference was significant(p<0.05).The most apoptosis of pancreatic epithelial cells in ducklings infected with pancreatitis-type DHAV occurred at 168h post-infection.The concentration of caspase-3 in liver of ducklings infected with hepatitis-type DHAV was significantly greater than that of infection with pancreatitis-type DHAV(p<0.05).While the concentration of caspase-3 in pancreas of ducklings infected with pancreatitis-type DHAV was significantly greater than that of infection with hepatitis-type DHAV(p<0.05).The ratio of AST/ALT in serum of ducklings infected with hepatitis-type DHAV was significantly greater than that of infection with pancreatitis-type DHAV(p<0.05).The concentration of AMYL and LIPA in serum of ducklings infected with pancreatitis-type DHAV was higher than that of infection with hepatitis-type DHAV,the difference was significant(p<0.05).Conclusion:Aftering infection with hepatitis-type DHAV,the apoptosis of hepatocytes are more obvious,both concentration of caspase-3 in liver and ratio of AST/ALT in serum were greater.Therefore,hepatocellular lesions are relatively serious.Aftering infection with pancreatitis-type DHAV,the apoptosis of pancreatic epithelial cells are more obvious,concentration of caspase-3 in pancreas,pancreatic amylase and lipase in serum were all greater.Therefore,the lesions of pancreas are relatively serious.There are differences in apoptosis,concentration of caspase-3,hepatic function and pancreatic function between infection with hepatitis-type DHAV and pancreatitis-type DHAV.
