| Rice blast,caused by Magnaporthe oryzae,is the major disease resulting in reduction of the yield and quality of rice.It’s wide seperated and has evolved the physiologial species that overcome the resistance of rice breed.It is a urgent promlem to prevent blast rice.The interaction between the Magnaporthe oryzae and rice is the model of researching interaction between fung and plants.The research that the early infection of the fungus and the adaptation mechanism in the lack of nitrogen can be used for control and prevention of rice blast,hence the study of the molecular mechanism of infection in rice and the pathogenic process has much important practical value and scientific significance.When we cloned and knocked out the MoGAP1,MoGAP2 and MoGAP3 genes,we found that the the pathogenicity ofΔMoGAP1 is weakest.So we chose MoGAP1to do the further research.This study has gotten the following results by researching the three permeases of absorbing amino acids in the nitrogen deficiency environment,the preliminary study of the General Amino Acid Permease MoGAP1,the function of MoGAP1 in the growth and reproduction:1.Via bioinformatic analysis of Mo GAP1,we got that the total gene length is2115bp,including 2 introns,210bp,encode 635 amino acids.2.The colony ofΔMoGAP1 is white,aerial hyphae are dense and thick,colony growth rate is lower than Y34,ΔMoGAP1 likes Y34 can form normal appressoria and penetration peg,the mycelial growth in onion epidermal cells,but it’s the number of conidias and the production of melanins are less than Y34.ΔMoGAP1 pathogenicity significantly decreased,even cann’t form disease spots when its conidias liquid inoculated on the rice leaf.3.Like Y34,theΔMoGAP1 has the same inhibition and growth tendency under the stress of adversity,and is not sensitive to H2O2 adversity.It is not sensitive to the stress of salt stress,suggesting that MoGAP1 gene has nothing to do with the osmotic pressure of the M.oryzae.The lack of sensitivity to Congo red and SDS suggests that the MoGAP1 is not on the cell wall.4.The experiment of recovering amino acids,in the lack of nitrogen source in MM-N and MM medium with nitrogen source the growth of MoGAP1 and Y34 is almost same,but the MM-N adding Ile,Lys,Thr,Met four kinds of amino acids,theΔMoGAP1 cann’t grow only in the Lys medium,and the Y34 can grow normally with Lys.We can conclude that MoGAP1 genes related to the absorption of Lys.The results show that MoGAP1 genes involved in vegetative growth of M.oryzae,the synthesis of melanin,the productive process of conidium,pathogenic process,amino acid absorption,and plays an important role in the pathogenic process. |
