In this paper,the effects of MeJA on the accumulation of the saponins and the expression of their biosynthetic key enzymes(FPS,SS,SE and CAS)in somatic embryos of P.notoginseng,were studied with the methods of the qRT-polymerase chain reaction(PCR)and HPLC.Also,the FPS gene plant expression vector was built,and that gene was transferred into tobacco plants using the mediation of Agrobacterium tumefaciens.Then these results were as follows:1.Among the 0?500?mol/L MeJA treatment on somatic embryos for 7 d,the highest total content of three kinds of ginseng saponins(Rg1,Rb1,Re)was 8.89 mg/g,treated by the 50?mol/L MeJA,and their accumulation efficiency also reached the maximum,0.4622 mg/g/d.With the increasing of MeJA concent,three kinds of ginseng saponins content gradually reduced;under the treatment of 500?mol/L MeJA,it achieved the lowest,3.72 mg/g;their accumulation efficiency also reached the minimum,0.0301 mg/g/d.The contents of Rg1,Re and Rb1 in the somatic embryos treated by 50 ?mol/L MeJA were the higher(0.96,1.65,1.65 mg/g)than those of other treamemnts;also their accumulation efficiency also reached the maximum value(0.0497,0.0858,0.3266 mg/g/d).With the increase of MeJA concentration,Rb1,Rg1 and Re contents were lower,in MeJA concentration to 500?mol/L reached the minimum value(0.16,0.28,and 0.28 mg/g).2.The relative expression of SS and FPS genes in 50 ?mol/L MeJA maximum when processing,and then decreased with increasing the concentration of MeJA;SE the relative expression of genes in the 250 ?mol/L MeJA maximum when processing.MeJA add,CAS gene expression and control significantly reduced.3.After induction MeJA,three kinds of ginseng total saponin content showed increased after decreased trend,then the MeJA processing after seven days maximum 8.89 mg/g.The original three glycol type Rg1 ginsenosides of ginseng and Re content increased gradually with the increase of the induction time,all in 10 days maximum(2.12 mg/g and 1.39 mg/g);Type and the original ginseng diol ginseng saponin content with the increase of the induction time of Rb1 showed a trend of increase after decreases first,and has been maintained at a higher level,induced maximum 6.29 mg/g after 7 d,6.45 times compared to the same period,the effect is very significant.4.FPS gene expression levels increase with the increase of the induction time,present time dependence,the characteristics of the MeJA induced 10 d when its expression level is 1.70 times.Compared with the control,SS gene and SE gene expression level increased significantly,when MeJA induced 7 d control of 4.95 and 2.45 times respectively.After the MeJA processing,CAS gene expression level and declined slightly.5.FPS gene plant expression vector was built;the gene was conformed into the tobacco plant by the PCR method,using the agrobacterium mediation.
