Chelidonium majus is a kind of very high prospects for the development of new medicinal plants.The main active ingredient of Chelidonium majus is chelerythrine.Chelerythrine is a benzo phenanthridine alkaloids and has anti-inflammatory,antifungal and other biological activities.In this paper,we studied the antifungal activity and antifungal mechanism of chelerythrine.Three kinds of fungi are obtained after separation and purification,18SrDNA molecule identified that they are Niqrospora sp,Asperqillus ruber,Chaetomium.The chelerythrine was added to the PDA medium to prepare a drug medium and the concentration are 5×10-3 mg/mL,1.O×10-2 mg/mL,1.5×10-2 mg/mL,2.0×10-2 mg/mL respectively,the antibacterial activity of chelerythrine is studied by using the isolated strains of Niqrospora sp,Asperqillus ruber.The results show that with the increase of the concentration of chelerythrine,the antibacterial effect is more obvious,and when the concentration is 2.0×10-2 mg/mL the inhibitory rate of chelerythrine against Niqrospora sp is 95%,and the inhibitory rate against Chaetomium is 92%.In this paper,the antibacterial mechanism of chelerythrine was studied from the following three aspects:(1)Effects of chelerythrine on cell wall of fungi.(2)Effects of chelerythrine on ROS content in spores.(3)Effects of chelerythrine on DNA.According to the results of optical microscopy and scanning electron microscopy,chelerythrine can induce folds of mycelium of Niqrospora sp.The chelerythrine can destroy mycelium and then produced cavities,the result can be observed under transmission electron microscope.Test the permeability of fungal cell walls,the absorbance of fungal extracellular fluid was significantly increased at 280 nm to confirm that chelerythrine had some damage to the cell wall of Niqrospora sp.After which the germination rate of spores in the drug medium was tested and the results showed the germination rate of spore decreased significantly under the action of chelerythrine.After staining with fluorescent dye H2DCF-DA,it could be clearly observed under the fluorescence microscope that chelerythrine could accumulate ROS inside the spores.Finally,agarose gel electrophoresis results show that chelerythrine can fragment fungal DNA.
