With the improvement of people's living standards,fruit have become everyone's essential food.However,fruit is one of the most common sensitized foods in recent years,peaches are frequently reported.Although five peach allergens have been identified in Europe,Pru p 3 is the main allergen causing severe allergic reactions to Chinese peach allergic patients.Different peach cultivars(nectarine)differ in allergenicity.To determine the Pru p 3 content in different peach cultivars,the Pru p 3 content of different peach cultivars were determined by the double monoclonal antibody-based sandwich ELISA method and the main results are as follows:1.We used little hydrochloric acid(20?l concentrated hydrochloric acid per 50ml peach extract)to treat the extraction of'Hujingmilu' to remove pectin in peach peel and the immunogenicity of Pru p 3 was not influenced as evaluated by indirect ELISA.The purified nPru p 3 was used as standard in in the following experiments.2.We tested the content of Pru p 3 in different peach mature stages.The results showed that the content of Pru p 3 was increased with the peach ripening among most of the peach cultivars,only a few were decreased.The Pru p 3 contents of different peach cultivars in different years(2014 and 2015)varied significantly from 0.29 to 11.62.For example,the ratio of Pru p 3 content of 'Hu Jing Mi Lu'in 2014 and 2015 is 11.62.3.We divided the tested peach accessions into four groups according to their potential allergencity:wild peaches,landraces,nectarines and peach cultivars.The results showed that the Pru p 3 content of peaches range from 6.43 to 311.81?g/g,the Pru p 3 content under 20?g/g could be regarded as hypoallergenic type.The Pru p 3 content of wild peaches was the lowest.about 17.30?g/g,followed by nectarines(35.78?g/g),landraces(81.34?g/g),and peach cultivars(93.83?g/g)was not significantly different.but much higher than that of nectarines and wild peaches.4.We also test the Pru p 3 content in epicarp,mesocarp and whole fruit.The results showed that the Pru p 3 was mainly accumulated in epicarp.The Pru p 3 content of mesocarp was much lower than that in epicarp.
