Tobacco mosaic virus(TMV)is an obligate intracellular parasite that has a relationship with the host cell environment,and it's completely dependent on the environment of host cell.The dependence requires the viral proteins and nucleic acids to change the function of host cells,therefore,it is extremely difficult to control the virus disease.Researchers have done a lots work to defence TMV disease for a long time,including biological agent,chemicals agent and genetic engineering etc,but there has been no significant progress.Therefore,the study of the molecular mechanism of the TMV infection host is conductive to prevent and treat the TMV disease of tobacco.In this study,i TRAQ technology was used to detect the changes of the tobacco proteme after TMV infected in different periods.At the same time,we analyzed the changes of metabolic pathway and screened the key proteins witch responding to TMV infection.Based on the key protein,the expression of the resistance gene of the number of disease spot,we detected the efficacy of control agents which is compounded for TMV infection.The results are as follows:1?Inoculating tobacco variety K326 with TMV,We used i TRAQ technology to detect the proteome expression profiles at different time points.144 differentially expressed proteins were identified at 12 hvs CK,of which 90 were up-regulated and 54 were down-regulated.There were 163 differential proteins in 24 hvs CK,among which 90 were up-regulated and 73 were down-regulated.In 36 hvs CK,the results showed 178 differential proteins,of which 97 up-regulated and 81 down-regulated.Gene Ontology(GO)was performed on the three groups of differentially expressed proteins: The results showed that these differential expressed proteins were mainly reflected in the fields of “electron carrier activity”,“catalytic activity” and “translocation regulation”,the differences in biological pathways were mainly reflected in the “metabolic process”,“cellular process”,“biological regulation process”.2?we performed KEGG enrichment analysis on the differential Proteome witch setted the limiting condition as P?0.05.When tobacco was inoculated with TMV for 12 h,the differentially expressed proteins induced by TMV were accumulated in “Biosynthesis of secondary metabolites”,“Terpenoid backbone biosynthesis”and “photosynthesis”.The differentially expressed proteins induced by inoculation with TMV for 24 h mainly accumulated in “starch and surcrose metabolism”,“Terenoid backbone biosynthesis” and “Photosynthesis”.At 36 hours after inoculation with TMV,the differential proteins induced by TMV manily accumulated in the “protein export”,“photosynthesis” and “porphyrin and chlorophyll metabolism”.It was shown that TMV had a significant effect on the photosynthetic system with 36 hours after invading tobacco.Cross-linking analysis of the differential proteins were respectively enriched in “RNA degradation”,“Porphyrin and chlorophyll metabolism” and “photosynthesis”.The results further demonstrate the effect of TMV infestation on the photosynthesic system.3?After we inoculated tobacco with TMV,the protein in the photosynthetic pathway changed,the number of up-regulated proteins gradually increased,and the number of down-regulated proteins gradually decreased.In porphyrin metabolism and chlorophyll metabolism pathways,the number of up-regulated proteins decreased and the number of down-regulated proteins increased.It shows that there is a compensatory expression of protein in tobacco during the invasion of TMV.By improving the expression levels of some proteins in porphyrin metabolism and chlorophyll metabolism,the damage of TMV to the photosynthetic system can be rescused to maintain the survival of plants.4?The results showed that application of preparation B before tobacco infection with TMV could increase defense enzyme activities and improve the expression of key genes and resistance genes involved in porphyrin metabolism and chlorophyll metabolism pathway,and enhance the resistance of tobacco plants to TMV.At the same time,Formulation B can reduce the damage of TMV to tobacco photosynthetic system.We used the half-leaf method to detect the disease phenotype of tobacco leaf after TMV infection,and also found that Formulation B had a significant effect on the control of tobacco mosaic disease.
