The nutritional quality of rice mainly depends on two aspects,including protein content and amino acid content.The content of digestible protein and the content of essential amino acids are particularly important for the nutritional improvement of rice.This study analyzes the nutrient components of rice storage protein,which can provide sufficient theoretical basis for the selection of rice varieties with high nutritional quality.Short feet nantes autotetraploid rice as material and the corresponding diploid as comparison,for the sake of researching different period of rice endosperm proteins of albumin,globulin,gliadin and gluten accumulation dynamics,this syudy uses the BCA protein concentration measurement technology to measure.For the sake of comparing gluten expression differences analysis in different materials during the same period and different periods of the same material,this study uses the gluten mass spectrometry analysis.The main results are as follows:1.The distribution of the storage proteins of the homologous tetraploid rice was basically the same as that of the diploid rice.Compared with the diploid rice,the content of the four main storage protein components was up-regulated and down-regulated.After 6 day pollination,albumin content of autotetraploid rice higher than diploid rice.After 7 day pollination,diploid albumin content higher than tetraploid and nearly flat after 20 day pollination.After pollination,the content of alcohol soluble protein was basically flat between 6 day and 9 day and the 9 day diploid content was higher than tetraploid after pollination.Gluten content is rising from 6 day to 9 day after pollination and the content decreased after 9 day pollination.After 12 day pollination,the content of gluten is increased and the content drop again after 15 day pollination.After 20 day pollination,gluten content obviously decreased than after 15 day pollination.The content of diploid and tetraploid glutamate was not very different,and the content of the diploid gluten was slightly higher than tetraploid in the early and the content of tetraploid was slightly higher in the late.The albumin-globulins content of rice and diploid rice was accumulated after 6 day pollination,and the content of alcohol soluble protein was accumulated after 9 day pollination.Gluten content began to decline after 9 day pollination,and after 12 day pollination continue to accumulate,15 day the accumulation amounts to the maximum,and then down after 20 day pollination.So after pollination,9 day 12 day,15 day and 20 day is an important point of gluten content accumulation.2.ITRAQ protein mass spectrum technology is used to analysis after pollination,10 day 15 day and 20 day short feet nantes autotetraploid and diploid endosperm glutelin differential expression,finding their differences in protein number a lot and a lot of protein has a variety of functions.(1)Short feet nantes autotetraploid rice compared with the corresponding diploid rice,after 10 day pollination with 125 differentially expressed proteins,after 15 day pollination differentially expressed proteins have 159,after 20 day pollination differentially expressed proteins have 78.There were 185 differentially expressed proteins in the 10 day,15 day and 20 day after pollination.The corresponding diploid rice had 372 differentially expressed proteins in the 10 day,15 day and 20 day after pollination.(2)GO analysis,after pollination,Short feet nantes autotetraploid rice up-regulated protein in rice in 10 day and 15 day were related to ribosomes,material metabolism and protein synthesis.After the double difference of rice protein more than the diploid rice species,20 d after pollination of raising rice protein decreased significantly,reduce the difference of protein is more associated with ribosomal proteins,and synthesis of ribosomal proteins in the differences in protein,is more than fifty percent of the differences in protein and rice metabolism,cell process and catalytic enzyme activity.The proteins that control cell tissue in differentially expressed proteins are upregulated proteins,and these differentially expressed proteins are related to grain phenotypes.Thus it can be seen that the metabolic capacity of pygmy nantes homologous tetraploid rice is stronger than that of diploid rice,and the grain size of tetraploid rice is larger than that of diploid rice.Autotetraploid and diploid rice rice protein expression of the same material trend similar in different periods,the synthesis of 20 d after pollination ribosomal protein are less than 10 d after pollination,and the differences in protein synthesis ribosomal proteins,most associated with rice physiological process.It can be seen that the metabolic capacity of rice decreases with the ripening of rice.(3)KEGG analysis,from the synthesis of lysine pathways found after pollination short feet nantes autotetraploid rice contains control the synthesis of lysine two proteins,these two kinds of protein is higher protein.Differences in protein synthesized by the control of lysine were B8AM24(2.6.1.83)B8ARJ0(4.3.3.7),15 d after pollination difference is most evident in the rice,pyridoxal phosphate binding protein B8AM24 control,synthetic amino transferase,the protein similar protein A0A0D3FHE9 A0A0E0NTW0 protein and protein,mainly in short feet nantes difference rice protein LL-diaminopimelate transaminase synthesis;Protein B8ARJ0 ACTS on lysine biosynthesis process through two amino valerate and synthesis of 4-hydroxy-tetrahydrodipicolinate synthase,the proteins of similar protein with protein Q7XM43,main role in short feet nantes difference of rice protein in the synthesis of 4-hydroxy-tetrahydrodipicolinate synthase.
