Petunia is an excellent landscaping plants,with unique shape and long flowering time.It have an important position in landscape garden.The reference genes can be expressed in all plant tissues.They are usually used as an internal reference genes to detect changes in the expression of target genes.However,there are few reports on their functions in plants.In this study,we try to identify the function of reference genes in petunia with the technology of VIGS.EF-1A,RPL10 and SAMDC play an important role in plants.The results of the study were as follows:EF-1A mRNA were present in all plant organs examined,including root,stem and leaf,by real-time PCR analysis.The expression level of EF-1A was highest in the stem.We successfully constructed VIGS vector pTRV2-EF-1A.VIGS-mediated EF-1A silencing resulted in dwarf plants.The plant height decreased and the leaf was seriously deformed.RPL10 mRNA were present in all plant organs,including root,stem and leaf by real-time PCR analysis.The expression level of EF-1A was highest in the stem.The expression of RPL10 was reduced under UV stress in petunia.We successfully constructed VIGS vector pTRV2-RPL10.Compared with the wild type plants,the plant height was decreased,the overall growth of the plant was decreased,and the number of stomata on the leaf epidermis was decreased.Compared with the wild type plants,there was no observable change in the plants of SAMDC silencing.
