Pinctada martensii(Dunker),an important nacre used to cultivate pearl.In the breeding process,due to changes in the environment and germplasm breeding a certain degradation,making a certain problem on pearl farming and fertility aspects.Since the 1980 s,P.martensi genetic breeding work began to receive attention of researchers.Shell color as the most apparent trait directly,shellfish shell color closely associated with economic traits,making shell color gradually become an important breeding trait.Since 2004,our group began P.martensi four kinds of shell color composite breeding populations studied,after seven years seven generations of breeding populations subculture,to 2011,has been significantly shell color and genetic stability The Shell Color Line.March 2012,for each shell color for the individual exists in each shell color heterogeneity,launched a large shell width Family Selection studies to differentiate the different economic traits pedigree,lay the foundation for supporting the establishment of the department.This study has constructed four different shell color shell width large pedigrees F1,using SSR markers of genetic diversity between each shell color family,using central composite design and the corresponding surface methodology temperature,salinity interaction on each Different shell color families affected P.martensi antioxidant power to verify the effect of shell color breeding,comparing different shell color P.martensi differences in the levels of resilience for healthy breeding P.martensi forecasts and genetic breeding environment to provide basic information.1.Microstatellite DNA technique was applid to estimate genetic diversity of four shell color families of Pinctada martensii.Useing six microsatellite loci,genetic parameters were calculated: the average effective numbers of alleles of the four families were from 1.297 to 1.819.The average expected heterozygosity and the average observed heterozygosity were 0.207~0.341 and 0.083~0.212,respectively.PIC values were between 0.257~0.442.Shannon's diversity was 0.366~0.616.The indices of Hardy-Weinberg were highly significant deviation in four color families.Fst values between four families were from 0.113~0.793.Genetic distance become larger,four families have lower genetic diversity.2.The combined effects of temperature and salinity on the total antioxidant capacity(T-AOC),malondialdehyde(MDA)in Pinctada martensii of the four shell colour families by using of central composite design and response surface method were studied,The range of temperature is 18?~34? and salinity is 20~40.This study established the equations of responses.The results showed as that:The model equations of the T-AOC towards temperature and salinity ware established,with R2 being 0.9613~0.9793,the establishment of the model is appropriate.The model equations of the MDA towards temperature and salinity ware established,with R2 being 0.9782~0.9990,the establishment of the model is appropriate.The coefficients of the model significance test and variance analysis showed that black shell color in temperature,salinity stress,the level of T-AOC,MDA concentration showed better resilience than the other three shell color.3.The combined effects of temperature and salinity on the GPx gene express in Pinctada martensii of the four shell colour families by using of central composite design and response surface method were studied,The range of temperature is 18?~34 ?and salinity is 20~40.This study established the equations of responses.The results showed as that:Liner effects and quadratic effects of temperature on the GPx gene express in four shell colors were highly significant;Liner effects of salinity on the GPx gene expres in block shell colors were significant,not significant in white,red and yellow shell colors;Quadratic effects of salinity on the GPx gene express in white shell colors were significant,highly significant in block,red and yellow shell colors;Interaction effect of temperature and salinity on the GPx gene express in block,red and yellow shell colors were significant,not significant in white shell color.The model equations of the GPx gene express towards temperature and salinity ware established,with R2 being 0.9713~0.9958,the establishment of the model is appropriate.The coefficients of the model significance test and variance analysis showed that black shell color in temperature,salinity stress,the level of GPx gene expresss showed better resilience than the other three shell color.
