Ammonia pollution of laying hens is a major problem that needs to be solved urgently.Previous studies have found that under the same conditions,the average daily ammonia emissions of romain's pink laying hens are about 22 percent lower than those of the farm's big seven.Among them,the difference of intestinal microbial community may be an important factor leading to the difference of ammonia production level in different kinds of laying hens.Therefore,this article through the 16 S r RNA,slice and scanning electron microscopy(sem),such as technology,by comparing the Roman powder and nongda no.7 the intestinal microflora in the differences in the distribution of time and space,look for Roman powder laying hens intestinal micro ecology systems help host low yield of ammonia in the performance of the special microbial communities,to analyze its mechanism of generation and engraftment,for further through regulating gut microbes lower layers of ammonia emissions and realize the sustainable development of laying hens industry to provide theoretical basis and technical support.This paper experiment mainly includes three parts,the contrast different ammonia production by 16 S r RNA sequencing technology level laying hens duodenum,jejunum and caecum intestinal flora difference,find the Roman powder and nongda 7 layers to produce ammonia difference key microbes;Trial by contrast 2,10,16 S r RNA sequencing technology,23,30,35 and 40,127,228,376 and 510 day-old Roman powder layers of duodenum,jejunum and colon and cecum contents and mucous membrane of bacteria types and differences,looking for the key microbes and engraftment regularity of key genes;Test three through associated with ammonia produced by physical and chemical index,slice,scanning electron microscopy(sem)analysis,to 2,40,127 and 376 day-old Roman powder laying hens the mucous membrane of the duodenum,jejunum and colon and cecum measurements and observations,search of intestinal development and the relationship between microbial engraftment.The main results are as follows:(1)The intestinal flora of Roman pink and nongda-7 laying hens was mainly composed of campylobacter,lactobacillus and clostridium(P<0.05).Among them,lactobacillus and clostridium are related to the body nitrogen metabolism of laying hens.(2)At the age of 2 days,duodenum,jejunum and colon have shown rich microbial community levels,and the species and quantity of caecum flora are extremely low.With the increase of day age,various types of gut microbes and quantity has increased,about 40 days of age remain stable,mucosal flora and contents flora changes have similarities,about 30 ~ 40 days of age,Roman powder laying hens to establish the function of nitrogen metabolism related genera,dominant fungi in 2~40 days of age,the remarkable increase of carbohydrate metabolism and nitrogen metabolism related index significantly increased(P < 0.05),so the Roman powder laying hens built around 40 days of age structure of intestinal flora.(3)The duodenum,jejunum and colon in 2 days of age has shown more perfect fluffy structure villus height were 377.96±15.34 nm and 239.84±16.23 nm and 247.52±11.09 nm,and cecum in 2~23 days of age,there is no apparent villi structure,40 days of age fluff structure development is complete,villus height of 460.86±31.49 nm.Above all,Roman powder laying hens in the 40 days of age structure,establish flora genera lactobacillus 2~40 days of age,such as nitrogen metabolism dominant fungi colonize the gut and cecum in the most significant,and increase the carbohydrate metabolism function,reduce the ammonia production.
