Tetraploid potato C88 is the mainly cultivar in southwest China.It has a very important position in the character of disease resistance,planting area and economic benefit.Although it has been confirmed that C88 has PVY and PLRV resistance,the related genotypes is not reported until this time,and potato virus disease causes the effect of yield lost and taste quality just after the late blight disease.In this study,969C88 self-inbred lines was used field phenotypes,DAS-ELISA assays,resistance gene linked molecular markes,cloning and sequencing,to analysis the genotypes of PVY and PLRV resistance genes,which are suitable for molecular marker for PVY and PLRV resiatance genes in self-inbred populations,and are benifit to linkage analysis between plant phnotypes and genotype linked to against PVY and PLRV.It provides a reference for the study on the antiviral and self-inbred population of C88.The main results are showed as follows:1.Phenotypes investigation in the field.The phenotypes investigation in the field showed that C88 had high resistance to PVY and PLRV.It is speculated that the resistance genotypes of PVY and PLRV were YYYy and RRrr,respectively.The corresponding separation ways were chromatid random segregation and chromosome random separation.2.DAS-ELISA detectionThe DAS-ELISA detection results of 969 strains of self-inbred population showed that there were very few individual plant carrying virus.Among of them,C88-69 and C88-1040 infected by PVY,47 individual plants were infected by PLRV.It is speculated that the resistance genotypes of PVY and PLRV were YYYy and RRrr,respectively.All the corresponding separation ways were complete equilibrium segregation way.3.DNA molecular markersIt is speculated that the genotypes of PVY resistance gene,Ryadg,has a genotype of YYYy and the corresponding separation way was complete equilibrium segregation way in self-inbred lines according to the detection by PVY linked molecular marker RYSC3.The PVY resistance gene,Rysto,has a genotype of YYYy and the corresponding separation way was chromatid random segregation by PVY linked molecular marker YES3-3B.The PVY resistance gene,Ry-fsto,also has the YYYy genotype,the corresponding separation ways were complete equilibrium segregation and chromatid random segregation of PVY linked molecular marker GP12271818 and GP122564,respectively.The PLRV resistance gene,PLRV.1,has a RRrr genotype and the corresponding separation way was complete equilibrium segregation by PLRV linked molecular marker Nl271164.The PLRV resistance gene,Rlretb,has a RRrr genotype and the corresponding separation ways were chromosome random separation and chromatid random segregation by PLRV linked molecular marker DMB32-11,1367-8a and C2-Atlg42990,respectively.The PVY resistance genes,Ryadgdg and Rysto,can be used to assisted selection linked molecular markers RYSC3 and YES3-3B in the self-inbred population of C88.The PLRV resistance genes,Rlretb,can be used to assisted selection linked molecular markers DMB32-11and 1367-8a in the self-inbred population of C88.In summary,in tetraploid potato C88,PVY and PLRV resistance genes genotypes were YYYy and RRrr,respectively.
