| Root-knot nematodes(RKNs),Meloidogyne spp.,are sedentary endoparasite nematodes of wide distribution.O ne of the most important types of plant parasitic nematodes attacking rice is the Meloidogyne graminicola,which causes serious yield losses to rice of the world.Numbers of effectors synthtised in the Esophageal gland of M.graminicola are secreted by the stylet into plant cells to promote their invasion,parasitism and develpoment during the infection,like other RKNs.Functional analysis of those effectors is helpful to understand the molecular pathogenesis of the nematodes and provides important theoretical basis for the development of new control strategies.In this paper,the function of M.graminicola effector Mg289 was furtherly studied by using in planta RNAi,subcellular localization,overexpression and yeast two-hybrid.The main results are as follows:1.The transgenic rices of RN Ai were obtained throngh agrobacterium-mediated genetic transformation approaches.We demonstated that the parasitic ability of nematodes was significantly inhibited through RN Ai.2.Over expressing of Mg289 in rice could improve the susceptibility to M.graminicola compared with the wild type rice.These results indicated that Mg289 serve as an important role during the nematode parasitism.3.In yeast two hybrid system,we screened three rice proteins,Os CB,Os Tha and Os HMA.The interaction between Mg289 and these three proteins were then further confirmed by contransformation into yeast.4.Transient expression of Mg289 and Os HMA in rice protoplasts showed that Mg289 and Os HMA were both localized in the cytoplasm and nucleus of rice cell.This result laid good reference for their interaction study.The results of this study provided a theoretical basis for further research of the role of the Mg289 effector in the parasitic process.Mg289 may serve as a reference for further finding a method to prevent root-knot nematode. |
