In early mammalian pregnancy,embryo implantation and decidualization are extremely important.Embryo implantation process requires the ability to implant the blastocyst and the uterus in the receiving state of the two processes simultaneously.Melatonin is an indole hormone secreted by the pineal gland,in vivo regulation of rhythm,antioxidant and anti-stress effect.Melatonin has a positive effec t on the embryonic development and in vitro blastocyst development of seasonal breeding animals such as cattle and sheep.However,the role and regulation of melatonin in implantation and decidualization of human and mouse embryos remains unclear,In this study,melatonin in human embryo implantation and decidualization of the regulation and role of the further analysis.We used real-time quantitative techniques and Western-blot and other experimental techniques to detect melatonin on endometrial epithelial cells and stromal cells in the receiving state and decidualization process of protein and m RN A levels.The results showed that melatonin could upregulate osteopontin(OPN)in Ishikawa cells and up-regulate OPN and Integrin-?3 in AN3 CA cells,and also increase the adhesion of trophoblast cells in endometrial epithelial cells.In vitro decidualized cell experiments,high concentrations of melatonin inhibit decidualization process,but in vivo melatonin synthesis of tryptophan and serotonin will promote decidualization.At the same time,in the cell proliferation experiment,1m M 5-hydroxytryptamine can inhibit the proliferation of endometrial stromal cells,but in decidualization will resist 1m M 5-hydroxytryptamine inhibit the proliferation of endometrial stromal cells.In the endometrial epithelial cells and stromal cells co-cultured melatonin on human decidualization marker IGFBP1 m RNA levels were significantly increased.In the process of decidualization,the two rate-limiting enzymes AAN AT and ASMT in the process of melatonin synthesis will change with the change of serotonin concentration,and in the endometrial epithelial cells,melatonin will activate fade Expression of Hormone Receptor 1 and Receptor 2.In the decidualization process,DNA damage and deiodinase changes,the treatment of endometrial epithelial cells with melatonin will inhibit the expression of DN A marker molecule p-Chk1,the same melatonin will promote the expression of Dio2 and Dio3.In summary,high concentrations of melatonin on endometrial epithelial cells have a role in promoting,but low concentrations of melatonin will promote the process of decidualization.So in the early pregnancy process melatonin play a supporting role.
