Based on the different pH,enzyme concentration,temperature,molar ratio of soybean under the condition of enzymatic hydrolysis of lipase in vitro test;and under different amount of enzyme,wheat,Bran,mixed feed for the substrate of xylanase enzyme test,this study investigatethe effects of enzymolytical feeds of lipase and xylanase in vitro,and 60d feeding trials were conducted in industrial culture system,to explore the effects of adding xylanase or lipase to GIFT,Oreochromis niloticus diets on growth performance,body indexes,whole-body composition,immune enzyme and antionxidant enzyme activities.Experiment 1:The Study of Enzymolytical Feeds of Lipase in VitroSoybean oil was chosen as the substrate of lipase to determine the optimal conditions of the lipase by measuring the production of acid value.The results shown that the optimal conditions for the lipase were:under 40?,pH 7.5,molar ration of water and soybean oil 40:1,for 7 hour.Under the optimal condition,degree of hydrolysis of soybean is 97.13%.Experiment 2:The Study of Enzymolytical Feeds of Xylanase in VitroWheat,bran and tilapia complete feeds were chosen as the substrates of sylanase to measure the production of the reducing sugar,there are four groups?0?5U/g?10U/g?20U/g?according to amount of xylanase.The results shown that xylanase enhanced the reducing sugar of feed.While xylanase enzymolysis wheat,wheat bran and tilapia complete feed,absorbable sugar enhanced160.58%,190.87%,205.39%,56.7%,82.29%,92.18%,47.89,62.44%,72.54%respectively.The effect of enxymolytical feeds of sylanase:wheat>wheat bran>tilapia complete feeds.Experiment 3:The Application of Lipase Supplementation in Diet for GIFT,Oreochromis niloticusIn the experiment,single-factor test design was adopted.GIFT,Oreochromis niloticus were randomly divided into four groups and fed on diets added with 0,3000U/kg(L3000),5000U/kg(L5000)and 8000U/kg(L8000)lipase for 60 days,each diet was fed to triplicate groups.Result shows that compared with control group,coefficient of feed of 0,3000U/kg,5000U/kg and 8000U/kg lipase addition group was decreased by 0.01%?P>0.05?,0.02%?P<0.05?,0.05%?P<0.05?,and the growth rate increased by 25.31%?P<0.05?,57.98%?P<0.05?,70.81%?P<0.05?;the addition of lipase significantly decreased the hepatic somatic index?HSL?,but there is no significant difference between treatment groups,and no significance difference was observed in viscera index?VSI?,relative fatness;the contents of crude ash,crude protein in whole-body composition were significantly improved by addition of 8000U/kg(L8000),meanwhile,no significant difference was observed in the contents of crude fat and humidity in whole-body composition among groups;LZM,AKP and ACP activity in the liver and serum of GIFT,Oreochromis niloticus fed with lipase diet promoted significantly,while there in no significant difference were observed in the activity of GSH-PX,T-AOC,T-SOD,T-AOC in liver and serum,and the most obvious results were observed in L8000 group.In conclusion,the addition of 8000U/kg might be helpful for improving the growth performance of GIFT,Oreochromis niloticus.Experiment 4:Application of Xylanase Supplementation in Diet for GIFT,Oreochromis niloticusThere were four groups of Gift,Oreochromis Niloticus which were fed with the basal diet supplemented without?X0?and with 500U/kg xylanase(X500),1000U/kg xylanase(X1000),2000U/kg xylanase(X2000)respectively,every diet was fed to triplicate groups.The experiment results show that the coefficient of feed of X0,X500,X1000 and X2000group was decreased by 0.11%?P>0.05?,0.25%?P<0.05?and 0.24%?P<0.05?,and the growth rate of 12.01%?P<0.05?,19.32%?P<0.05?and 15.91%?P<0.05?,fish fed the diet containing 1000U/kg lipase has the lowest FCR and highest coefficient of feed;HSL has decreased 0.32%?P<0.05?,0.76%?P<0.05?,0.38%?P<0.05?respectively,even there is obvious difference between X1000 and X500,X2000,but no significance difference was observed in viscera index?VSI?;the addition of xylanase significantly decreased the hepatic somatic index?HSL?,and there is significant difference between X1000 and X500,X2000,but no significance difference was observed in viscera index?VSI?,relative fatness;the contents of humidity in whole-body composition were significantly improved by addition of xylanase,but there is no obvious difference between treatment groups,and significant difference was observed in the contents of crude fat,ash and protein in whole-body composition among groups,X1000 group is best obvious;GSH-PX,T-SOD,AKP,ACP activity in the liver and serum and T-AOC activity in liver were promoted significantly,while no significantly difference was observed in the activity of CAT in liver and serum and T-AOC in serum,the most obvious results were observed in X1000 group.Therefore,the addition of 1000U/kg might be helpful for improving the growth performance of GIFT,Oreochromis niloticus.
