The normal bacterial flora structure in intestinal of piglets would change when PED and intestinal bacterial flora is very important for maintaining the health of piglets.Taking intestinal contents of PED and 15 day-old healthy piglets as research objects,study the bacterial flora structure differences in the feces and intestinal contents of PED and healthy piglets,explore the general changing rulers of intestinal bacterial flora induced by PED.ELISA test conducted firstly,and then analyzed the differences of bacterial flora structure in the feces and intestinal contents of PED and healthy piglets by the technology of bacteria-selective culture and PCR-DGGE.ELISA test results: ELISA results were PEDV negative in 10 tests of feces from healthy piglets could be as control group.There were 8 PEDV positive results in 10 samples of inspected feces infected by PEDV,which could be used as to study the bacterial flora structure of feces from PED piglets.PEDV results of both large and small intestine contents were positive tested by 4 suspected samples infected by PEDV,which could be used as to study the bacterial flora structure of intestine contents from PED piglets.In the trial of faces bacterial selective-culture count(Agar,MacConkey,Pfizer,BL and MRS etc.)from PED and health piglets,contrasted with the healthy group,the total bacterial of PED group decreased and was not different,lactobacillus and bifidobacterium quantities of PED group decreased differently(P<0.05).They both raised on coliform and enterococcus counts but no significant difference.These results shown that the intestinal bacterial flora will altered in which the probiotics-lactobacillus and bifidobacterium decreased,and pathogens-coliform and enterococcus increased when PED.Analyzing bacterial flora structure in the feces and intestinal contents of PED and healthy piglets show that there are generalities between DGGE finger-print of PED and healthy group.Analyzed the generalities and contrasted the differences.From DGGE finger-print of faces and intestinal contents,the healthy group were more complex than PED group.The bands number on DGGE finger-print of health group were more than PED group indicated that PED decreased the number of original dominant bacterial floras.In PED group,the number of pathogenic bacterium such as enterococcus increased and probiotics,such as lactobacillus decreased,and other pathogenic bacterium found also.These results demonstrated that PED caused alteration of intestinal flora and changes of bacterial flora of piglets.
