Objective: 1?To observe the external morphological characters of the mites Geckobia which is the ectozoa of gecko(from Shandong),and analyze its phylogenetic relationships based on 18 SrRNA genes and mitochondrial cytochrome oxidase subunit I gene(COX-1).2?To campare the composition and diversity of bacteria in mites Geckobia and the host.Methods: 1?Mites were collected from the skin of gecko which is from Shandong province,and morphological observation of the mites was performed by super-depth microscopy and scanning electron microscopy.The 18 S rRNA,COI genes from the genomes of Geckobia were amplified by PCR and sequenced for homology analysis.2 ? The metagenomic 16 S rDNA profiling of bacteria were sequenced by Hiseq PE250 sequencing platform to describe the bacterial community in mites Geckobia,to evaluate bacterial population diversity and to identify the predominant microorganisms and distribution of pathogens.Results: 1?The mite in experiments is flat and wide in shape,the average size is 450±20?m×470±20?m,the idiosoma is in bright red with four pairs of leg,gnathosoma is very small.Body surface with numerous silver white,thick,plumose setae.Propodonotal shield is 170?m long and 45?m wide.The morphological characters of experimental mites are highly similar with the species Geckobia Gerrhopygus.sp,described by Monika Fajfer.2?The length of the 18 S and was 307 bp,which had a high homology of 98.0% compared to Geckobia(NO.AF142114).The length of COI gene sequences was 703 bp,which had a lower homology in 75.79% compared to Pterygosoma(NO.KT962103).3?The total amount of Tags obtained from experimental mites is 60071 and 62242 from host.Conclusion:1?This study detected the morphology and phylogenetic relationship of Geckobia,and this is the first time for Geckobia to be reported in China,which means it probably is a new species of mites of genus Geckobia in Asia.2?There are some differences in the composition and predominant species of bacteria between Geckobia and the skin of gecko,some unique bacteria may be obtained in Geckobia's free life stage,and the composition of these bacteria need to be further explored.
