Two cultivars including zhongmiansuo-12 and Guokang-1 with higher and lower waterlogging resistance,respectively,were used as materials in this paper.Firstly,the protein expression of cotton root under waterlogging stress were analyzed by iTRAQ technology,and subsequent bioinformation analysis including GO analysis and Pathway analysis were carried out to identify key genes related to waterlogging in cotton.Then,the expression profile of these candidate genes at mRNA level were validated by real-time quantitative PCR.The main results are as follows:1.A total of 407 proteins were identified in Zhongmiansuo 12 and Guokang 1,two cotton cultivar with different resistance ability to waterlogging by iTRAQ quantitative proteomics technology.2.GO analysis showed that the differentially expressed proteins were identified in the biological process of cotton,including about 25% of these proteins are associated with metabolic processes,approximately 24% of them are involved in cellular processes,and most importantly,about 12% of the proteins are associated with cellular response to stimulus;In terms of the differential protein involved in cellular component,cell related protein accounted for about 30%,and cellular components related protein accounted for about 29%,and organelles related protein accounted for about 22%,and organelles components related protein accounted for about 10%;These protein were divided into 3 different categories accoding to their molecular function,including about 48% of identified proteins with catalytic activity,about 5%t with transport activityr,and about 44% with the binding activity.3.Six differentially expressed proteins were found in TCA pathway by Pathway analysis,including 2 up-regulated proteins and 3 down-regulated proteins.Up-regulated proteins are isocitrate dehydrogenase and malate dehydrogenase,respectively.Down-regulated proteins comprise dihydrolipoamide dehydrogenase,isocitrate dehydrogenase and ATP-citrate lyase.4.The expression profile of 30 genes related to waterlogging resistance was analyzed by qRT-PCR analysist.The expression levels of four genes including MDH1,At4g29360,Ubqln1,PME7 were up-regulated in Zhongmiansuo 12,the cotton cultiar with higher resistance to waterlogging.
