Eriocheir sinensis is one of the important economic crabs in China.It has the characteristics of reproductive migration and is an in vitro fertilization species.Various metal ions in the sea water are of great significance to the sperm acrosome reaction and fertilization.The K+potential is the closest to the cell resting potential on the cell membrane,so its ion channel plays an important role in maintaining the cell resting potential.The Slack channel,as a potassium channel activated by sodium,plays an important role in balancing the K~+concentration of the sperm membrane in the sea water with a large amount of Na~+during the reproductive migration of the crabs.In this experiment,the antibody of the Slack channel of Chinese mitten crab was prepared to provide a basis for the study of its expression and function in the Chinese mitten crab sperm cells.This experiment through analysis laboratory sinensis testis transcriptome data,and bioinformatics analysis,determine the preparation of antibody immune the original sequence.The expression vector of Slo2.2 was successfully constructed by genetic recombination technique.The histidine label on the expression vector was used to purify the recombinant protein and obtain the immunogen.With the ELISA experiment,the antibody titer of 5laboratory mice were detected to be more than 1:10000.Using Western Blot technique to verify the experiment of the preparation of antibody can effectively combined with the immunogen,antibody is available,and in the sperm of eriocheir sinensis and testis were Slo2.2 exists...
