Ruditapes philippinarum is an important economic value of marine shellfish,in recent years,due to non-standard farming methods and overfishing and other factors,as a result,the germplasm quality of ruditapes philippinarum began to degenerate,utilize germplasm resources for science,It is necessary to analyze the genetic structure of ruditapes philippinarum,learn about the differences in clam groups in the philippines,and establish a method of distinguishing between different populations.A total of 66 microsatellite primers were screened in this experiment,among them,43 pairs of primers had specific amplified bands in the six experimental groups,amplification success rate of 65.15%,among them,20 pairs of microsatellite primers have the characteristics of high interspecific compatibility and good polymorphism.A total of 300 samples from 6 groups of ruditapes philippinarum were amplified and analyzed,atotal of 518 alleles were detected.In 6 groups,The average number of alleles(Na)of zebra clams,Fujian,Guangxi,Yingkou,Laizhou and Tianjin populations were 4.75-5.7.The average expected heterozygosities(He)were 0.7011-0.7539.The highest value of 20 loci was detected in Tianjin(0.8660).While in Guangxi(0.4446)the lowest value.The PIC value of microsatellite loci was between 0.3922 and 0.8444.The largest PIC of RPg15608 in Shandong Laizhou population was 0.8444.The PIC value of RPg16975 in Tianjin population is the smallest,which is 0.3922.In Laizhou,Shandong,Yingkou,Fujian,clam groups in 20 sites showed a high degree of polymorphism,the percentage of highly polymorphic loci was 100%.The percentage of highly polymorphic loci in Guangxi population was 95%.The percentage of highly polymorphic loci in Tianjin population is 90%.PIC values of 2 markers(RPg15429,RPg15608)in all six populations were higher than 0.7,these two markers can be used as a valid genetic marker.The average polymorphic information content(PIC)was 0.6421-0.7068,respectively.The genetic diversity of the six groups was Shandong Laizhou> Zebra clam> Fujian> Guangxi> Tianjin> Yingkou.Most of them exist significant or very significant deviation from the equilibrium of Havan,indicating that microsatellite primers may have invalid alleles in the population,at the same time,it also indicated that the six groups tested in this paper are likely to have serious inbreeding.Nei's similarity and genetic distance between six populations in the Philippines were calculated,Nei's similarity coefficient was 0.6591-0.8472,the genetic distance was 01659-1.4169,Fujian and Yingkou had the highest similarity coefficient of 0.8472,while the genetic similarity between Fujian and Tianjin was the lowest,0.6591.In this study,we combined the principles of molecular biology and programming to develop and produce two software models: One is the web version of the software;the other is the more complicated algorithms programmed to produce separate program software.Web version of the software has the advantage of simple operation,do not need to download,there are networks to operate;The disadvantage is that in the absence of the network can't operate,and require MATLAB to change the fileformat,there are limitations.Independent identification software,the user can download without network operation,and does not require MATLAB to change the format of the allele frequency database,It does not need to run under the network state,enter the database directly in EXCEL software,Finally imported into the program to get the similarity matrix immediately,the operation is simple and convenient,accurate to 6 after the decimal point,the result is more accurate.
