Study On Germplasm Characteristics And Genetic Diversity Of Wumengfeng Chicken

Wumengfeng chicken'some performace(growth performance,reproductive performance,slaughter performance,meat quality indices)were studied in this study,at the same time,the technologies of mtDNA D-Loop region and microsatellite DNA marker were used to research for genetic background and genetic diversity.Research results were as follows:1.The body of Wumengfeng chicken was great,and the cock was better than the hen on entirety somatotype;Wumengfeng chicken' age at first laying was 181~210 days,the egg shape index was 1.33±0.06,the Haugh unit was 65.88±3.56,the fertility rate of breeding eggs and hatchability of fertile eggs reached more than 90%,it was one special native chicken with meat performance mainly;Results of the butchery rate,half-net chamber rate and net chamber rate were 89.37% and 91.82%,80.82% and 78.11%,68.7% and 65.14% respectively in cock and hen,the Wumengfeng Chicken have the characteristics of good carcass capability,and it was in high quality;Results of cock and hen about the percent of content of unsaturated fatty acid,zinc rich ability,selenium content were 61.74% and 64.19%,34.20mg/kg and 27.80mg/kg,0.70mg/kg and 0.70mg/kg,respectively;T\|test showed that the difference between the cock and hen was insignificant(P<0.05),the meat has high nutritive value.2.Studied on mtDNA D-Loop region showed Wumengfeng chicken'sequences were located in 1227~1229bp,the nucleotide level of A,G,C,T were 26.4~26.7%,13.2~13.4%,26.2~26.8%,33.5~33.9% respectly,content of A+T was 59.77~60.47%,content of G+C was 39.53~40.23%,content of A+T was above content of G+C;Analysis showed that there were 38 variable sites,including 26 T-C conversion,8 A-G conversion,2 A-C transversion,2 insertion-deletion(located in 12 bp,855bp);The diversity index of haplotype diversity,nucleotide diversity,the average degree of nucleotide,haplotype ratio were 0.90,0.01,8.68 and 32.50%,respectively.Genetic Diversity of Wumengfeng chicken was rich.NJ tree done by MEGA6.0 showed that there were more maternal origin.3.There were showed mean number of alleles,effective number of alleles,mean SI respectively were 83,4.3684,3.0939,1.2303,by microsatellite DNA marker.Hardy-Weinberg balance showed the loci number was 7,and the others were in an unbalanced state;Population genetic differentiation coefficient was between 0.5270~0.2987,indicated 21.10% variation was from gene flow;The gene flow was 0.5586~2.2867;Genetic distances and phylogenic tree showed that the genetic diatances was 0.3265~0.4863,and Wumengfeng chicken has closer relationship with Gaojiao chicken,Aijiao chicken,a near relationship with Weining chicken,a farthest relationship with Zhuxiang chicken,Baiyi black chicken.
