Copper?Cu?is one of the important metal elements in crustaceans.Containing Cu protein or containing Cu enzyme plays an important physiological function in the process of electron transfer,oxygen transport,oxidation reduction of substrate and regulation of material absorption.Excessive or too low Cu in a living organism or environment can cause stress or deficiency in the organism.In order to explore the physiological effects of Cu on the crustaceans,Litopenaeus vannamei was used as main experiment object.Chronic enrichment was used to determine the effect of copper in different concentrations or dissolved form during 56 days on growth,immune and metabolic function substances in Litopenaeus vannamei.In addition,the content of muscle Cu in each group was supplemented to study the enrichment of Cu and the problem of food safety.The experiments were divided into five groups:one control group without Cu addition,two groups of different dosage of complexing Cu group and Cu addition group with which matched the complex Cu group released Cu.The LC6911 type corrosion-resistant alloy Cu was used as a complex Cu source,using 0.15m2/M3 and 0.3m2/M3respectively,and the other two groups added matched amount CuSO4·5H2O as Cu2+source.The following conclusions were drawn:1.In the water of non-cultured,the experiments used LC6911 type corrosion-resistant alloy Cu tiled with 2m3 aquarium barrel bottom.The total Cureleased by copper plate were directly proportional with those area.2.Immune proteins'including CP,POX,MT activities was improved under two concentration gradients in experimental group,and which was significantly higher than control group?P<0.05?.3.The enzyme activity of LZM in shrimp hemolymph was found in the experimental group of two different dissolved states.The results showed significant difference among control group and others at the end of the experiment?P<0.05?.There was no significant difference between the two types of dissolved Cu and the different additive levels.And there was no significant difference in the LZM activity between the experimental group and the control group in gill tissue either?P<0.05?.But the activity of LZM in Cu2+group decreased significantly?P<0.05?.4.The long-term growth data showed that the growth effect of the experimental groups were way better than control group,and the growth effect of the 0.15m2Cu-plate/M3 group was the best.The enhancement of the immune activity and growth of Litopenaeus vannamei by using complexing Cu in water is more prominent than Cu2+,meanwhile,using 0.3m2copper plate/M3 in seawater has lower long-term toxicity effects on Litopenaeus vannamei's immune system.During eight weeks of experiments,Cu2+-ATPase,Ca2+-Mg2+-ATPase,CuZn-SOD,and Cu content were detected in different tissues.Results:The presence of two forms of Cu dissolved waters affected the Cu Content level in L.vannamei muscle tissue,but the content levels did not increase as time and Cu levels up,it decreased at different time nodes.Cu2+-ATPase activities in gills and hepatopancreas is directly proportional to Cu content level.The activities of Ca2+-Mg2+-ATPase in the two tissues showed a tendency of rising first and then decreasing.The activities were positive in the early period and were inversely proportional to the extension as the time goes.The activities of CuZn-SOD in the gills,hepatopancreas and hemolymph tissues showed a different trend of change,and more undulating and undulating changes.The experiment shows that within the scope of this experiment,different dissolved Cu and Cu contents will not cause Cu accumulated too much.A certain amount of Cu can promote the metabolic growth of L.vannamei,but over time will inhibit the absorption and utilization of calcium and magnesium in L.vannamei.Under the experimental conditions,the dissolving form of Cu in water in gills and hepatopancreas showed that the effect of Cu's form on enzyme activity was less than that of Cu content levels.
