Mongolian horse is one of the five type of horse breeds in China.Under the long-term natural and artificial selection,the Mongolian horse adapts to extreme climate,the resistance of forage,strong disease resistance and endurance characteristics.Compared with other horse breeds,the endurance performance is extremely prominent in the Mongolian horses.The research on horse kinematics are mainly focused on Thoroughbred,however,the mechanism of kinematics of the Mongolian horse is not clear.In this study,six Mongolian horses were used as the experimental subjects,and the skeletal muscle of the the Mongolian horse was performed by Miseq high-throughput sequencing technology for sequencing and bioinformatics analysis.We performed a transcriptome-wide analysis of skeletal muscle tissue in six Mongolian horses before and after a bout of high-density exercise(20 mins).We built transcriptome library using Miseq high-throughput sequencing technologies.Then the obtained etranscriptome sequences aligned to horse reference genome.Finally,analysis of differentially expressed genes,GO enrichment and KEGG pathway were perfomed.The results showed that the transcriptome data obtained 25,367 SNP sites,with the highest number of SNPS on ECA 1 and ECA 11 in the Mongolian horses.In addition,four kinds of splicing types were found,among which the exon jump type was the most.The number of variable splicing is obviously higher than before operation.Before and after the exercise,205 significant difference expression genes were detected in the two groups(P < 0.05).120 genes were up-regulated after the exercise,and 85 genes were down-regulated before the exercise.The results of GO functional enrichment analysis showed that 77% of differentially expressed genes were enriched in biological processes and 7.7% enriched in cell components,and 15.3% enriched in molecular function.Among them,enriched in the biological process,including the PDK4,CYR61,TNNC1 gene;enriched on the molecular functional pathway,including the TRIM63,LP1N1 and the MYL3 genes;and enriched on the cellular pathway,including ACTN,VEGFA,EGR2,ITGAV genes.It is speculated that the short-term high-load movement caused the body to undergo changes in the metabolic mechanisms such as Circulatory system process,nucleic acid binding transcription factor activity and enzyme binding.KEGG pathway analysis identified the main enrichment of differentially expressed genes in lipid metabolism,signal trancription,cell communication,endocrine system,cardiovascular disease,and other related pathways;And EGR2,MHC1,MYL3,ITGAV,TR1M63,LP1N1,ACTN,VEGFA,PDK4,CYR61 were screened,and these genes may be involved in regulating the Mongolian horse short-term high load exercise physiological process.
