| Chrysoperla sinica(Tjeder)is an important predator in farmland ecosystem,which is widely distributed in China and has a large population.In order to explore the physiological function and ecological significance of endophytic bacteria to the host inset,in order to know the distribution of chrysoperla sinica(Tjeder)at different stages of development,whether the microbial composition in its body will change with the developmental period,and whether there is a difference in the composition of the microorganism in the body of the different species of Chrysopa.The understand the microorganisms in the body is of great significance for a host of reproductive and developmental and environmental adaptability,and provides a theoretical basis for the artificial cultivation of the chrysoperla sinica(Tjeder).This research mainly is to use the 16 SrDNA gene sequences,build 16 SrDNA clone library,studied the field of Chrysoperla sinica(Tjeder)in microbial community structure,and Compare the different genders of Chrysoperla sinica(Tjeder)and different green lawewing.The Miseq high-throughput sequencing was used to analysis and compare the bacterial diversity and community composition of different developmental stages of Chrysoperla sinica(Tjeder),obtain the main results are as follows:1.The result indicated that the bacterial could be divided into three major groups respectively Gamma Proteobacteria,Alpha proteobacteria and Beta Proteobacteria.The chrysoperla sinica(Tjeder)adults symbionts is mainly composed of Wolbachia sp.,Rickettsia sp.,Enterobacter sp.,Uncultured Methyloversatilis sp.,Serratieae sp.,Pantoea sp and Erwinia sp classify by genus Only Cosenzaea sp in famale,Acetobacter sp.,Asaia sp.,Rosenbergiella sp.,Citrobacter sp exists in males.The dominate symbionts in chrysoperla sinica(Tjeder)adults is Wolbachia pipientis,and account for 23.3 % in males and 18.4% in females.Various bacteria composition were in the chrysoperla sinica(Tjeder),there are individual differences between male and female groups.2.By building 16 SrRNA clone library,the chrysopa pallens adults symbionts is mainly composed of Acinetobacter sp.,Pantoea sp.,Uncultured Enterobacter sp.,klebsiella sp.,and Escherichia coli.Both chrysoperla sinica(Tjeder)and chrysopa pallens have Pantoea sp and Enterobacter sp.We selected the subdominant Enterobacter sp from chrysoperla sinica(Tjeder)and chrysopa pallens and analyzed the homologous analysis with the results of sequencing,which had high homology in of chrysoperla sinica(Tjeder)and chrysopa pallens.3.By extracting the genomic DNA samples from seven different developmental stages,overall 123 optimized reads were obtained from these samples based on high-throughput sequencing of the V3-V4 region of the 16 SrRNA gene.Bacterial species detected in these samples covered 8phylum、class 16、Order 29、family 62、genus 89.Analysis showed thatthe bacterial diversity was very high in these samples,and there were differences among different samples.The distribution characteristics of the dominant bacteria showed a dynamic change model in different developmental stages of Chrysoperla sinica(Tjeder).Taxonomic assignment analysis indicated that Proteobacteria were the main bacteria,and their relative abundance accounted for 29% to 98.41% at phylum.Samples at different growth stages of bacteria abundance in the top genus,Rickettsia was a bacteria at every growth and development stage,so preliminary speculates that the endophytic bacteria can be passed through vertical transmission way. |
