| Wheat lesion mimic mutant LF2010 in EMS mutation of common wheat flower a genetic stable breeding culture lines from H261 in the offspring as the research material,through the analysis of different materials of the genetic background of authenticity of identification,and the use of gene targeting strategies and different to this new class of lm3 gene mutation in wheat disease Gene localization.The main results are as follows:1.Analysis of genetic models: a lesion mimic mutant mutant LF2010 and normal strains of spring Chinese hybridization,respectively.The F1 and F2 groups and F2:3 pedigree genetic model analysis,the results showed that dot LF2010 traits in segregating populations of hybrid LF2010 and Chinese spring by 1 pairs of recessive nuclear gene control.2.The identification of mutant genetic background: by wheat anther culture lines H261 after three EMS mutants were obtained for authenticity identification.The genetic background of LF2010、LF2099 and their parent H261 is highly consistent with H261,while LF2100 is the natural outcrossing or mechanical hybrid produced "false mutant" conclusion,for subsequent experiments excluding the uncertain factors.3.The location of lm3 :using BSA,wheat 660 K gene chip combined with BSA gene localization strategy,and using KASP high-throughput genotyping platform,achieve the target gene localization region primer encryption work,finally got 24 pairs of KASP primers and target gene linkage,the target gene was mapped on chromosome 6BL of wheat 6B03 and 6B40 2.36 physical distance,and found 58 candidate genes. |
