The purpose of this experiment was to study the effects of soy isoflavones supplementation on the feed intake,weight,blood reproductive hormone concentration,antioxidant and semen quality of sannen bucks,and to explore the effect of 0.05 mM and 0.1 mM soy isoflavones on the preservation of semen at normal and low temperature in the semen preservation diluents of dairy goats.The test selected twelve 2-3 years old bucks with good semen quality and weight(72.5± 6.33)kg.These bucks was divided into two groups according to the genetic pedigree data and weight randomly: control group(no supplementation of soy isoflavone,n=6),soy isoflavone addition group(each supplemented soy isoflavone 100 mg/days,n=6).The test period was 4 weeks,and the test period was 10 weeks;0,0.05,0.1 mM isoflavones were added to the semen preservation diluent.The effective survival time,vitality,plasma membrane integrity,acrosome integrity,T-AOC,ROS,MDA,and other semen quality indexes were detected at normal temperature and low temperature.The results are as follows:1.Compared with the control group,the supplemental soy isoflavone could significantly increase the intake and feed intake of the SAM goats(P<0.05),and had no significant effect on the weight of the goats and serum testosterone(P>0.05),significantly decreased the level of serum estradiol(P< 0.05),and significantly improved the serum SOD and T-AOC.Level decreased MDA level(P<0.05),but had no significant effect on GSH-Px level(P>0.05).2.Supplementation of soy isoflavones in diet did not affect ejaculatory quantity,semen pH,sperm viability,vitality and acrosome integrity(P>0.05),but significantly improved the integrity of plasma membrane(P<0.05).3.In the preservation test of semen at normal temperature,the addition of soy isoflavones significantly increased the vitality and plasma membrane integrity(P<0.05)after long preservation of sperm,increased the level of T-AOC and ROS,reduced the level of MDA(P<0.05),but had no significant effect on the effective sperm survival time and the acrosome integrity(P>0.05),0.1 M M has the best effect.4.In the cryopreservation test of semen,the addition of soy isoflavones significantly improved the effective survival time,vitality,plasma membrane integrity and acrosome integrity(P<0.05),increased the level of T-AOC,reduced the level of MDA and ROS(P>0.05),and the optimum concentration was 0.1 mM.To sum up,the supplementation of soy isoflavones in the diet could significantly increase the feed intake and antioxidant capacity of the goats,but it could not significantly improve the quality and reproductive performance of the semen,and could significantly improve the preservation of the semen at room temperature and low temperature.The optimum concentration in the diluent was 0.1 mM.
