White scour of piglets is a digestive disease that seriously affects the pig industry and is mainly caused by E.coli.Wumei granules are composed of ebony,berberine,turmeric,wolfberry fruit,and persimmon.It has the functions of clearing heat and detoxifying,and astringent intestines to stop diarrhea.In this study,Wumei Granules were subjected to in vitro antibacterial test,acute toxicity test,subchronic toxicity test and target animal safety test.The following results were obtained.1.In vitro bacteriostatic test of Wumei GranulesDetermination of the diameter of the inhibition zone: The disc diffusion method was used to measure the diameter of the inhibition zone.The results showed that different concentrations of Wumei Granules have antibacterial effect.Determination of MIC and MBC: Determination of the minimum inhibitory concentration and the minimum bactericidal concentration of Wumei granules by the tube dilution method.The results showed that the MIC was 110 mg/mL and the MBC was 220 mg/mL.2.Maximum dose determinationTwenty female and half male mice were randomly divided into 2 groups.The first group was fed with 40.0g/kg of ebony granules and the second group was administrated with 0.6mL/20 g of normal saline.After 7d,the mice were observed for death and the maximum tolerated multiple was calculated.The results showed that the maximum dose of Wumei Granules to mice was 40.0g/kg,and the maximum tolerated multiple was 150 times.3.Sub-chronic toxicity test of Wumei Granules80 SD rats were divided into males and females(fasting before the test).Female rats and male rats were divided into 4 groups with 10 rats in each group,40g/kg in high dose group,20g/kg in low dose group,and low dose.Dose group 10 g/kg gavage ebony granules and gavage 20 mL/kg physiological saline,fed once a day,continuous gavage for 28 days.During the experiment,the rats were observed for general symptoms such as feeding,drinking,fecal shape,and mental activity.Rat body weights were measured and recorded before and at the 7th,14 th,21st,and 28 th days of the experiment,respectively.After the end of the experiment,the organ was taken to observe the pathological changes,and the organ coefficient was calculated and the pathological tissue section was observed.Finally,the hematological parameters and blood biochemical indexes were determined.The results showed that,during 28 days,the rats had normal feeding habits,no abnormal drinking water,fecal shape,well-groomed eyes and eyes,and active spirit.All rats did not die,their organs did not have lesions,and their body weight increased.In the heavy effect,the organ coefficient,hematology index,and biochemical index were not significantly different from that of normal saline in the same sex group(P>0.5).No organ lesions were found in the organs of rats.4.Wumei Particle Target Animal Safety TestHealthy piglets 40,randomly divided into 4 groups,each group of10,10g/kg,30g/kg,50g/kg of ebony granule group and the control group(drug),trial for 14 d,before and after the test check the nervous system,digestive system,respiratory frequency and heart rate.Anticoagulant blood was collected at 1d before and 1d,3d,7d and 14 d after administration,respectively,for examination of hematological and biochemical indicators.The results showed that the mental condition of the piglets was good,the feces and urine were not abnormal,the breathing frequency and heart rate were stable and within the normal range.Before and after the administration of the medicine,the hematology index,hemodynamics index of the experimental group and the control group showed no significant difference(P> 0.5),indicating that wumei granule was safe for the clinical use of piglets.
