As a new type of probiotics,Clostridium butyricum is thermotolerant,acid tolerant and resistant to a series of antibiotics.Adding of Clostridium butyricum preparation in livestock and poultry feed is significant to reducing the overuse of antibiotics,and the drug tolerance of animal bacteria,then protecting animal health in the present feed industry.Clostridium butyricum SHYY088 was provided by Wuhan Sunhy Company.This paper mainly focuses the mycelia content of Clostridium butyricum.According to the influencing factors such as the composition of the Clostridium butyricum medium,the optimum pH and the culture temperature,the seed culture process and the fermentation process are optimized.And also study selection drying auxiliary agents in spray-drying process.The seed culture condition of Clostridium butyricum was optimized via liquid static culturing method and the medium formula and culturing parameters were determined.Based on the optimized seed preparation,the fermentation condition for Clostridium butyricum was optimized further.Temperature of 32 ~oC,pH of 6.5,maltodextrin as carbon source and yeast autolysis powder and yeast extract as compound nitrogen source,0.3%sodium phosphate as buffering system was found to be the optimal,which can give a viable count of Clostridium butyricum as high as1.39×10~9 cfu/mL.In a addition,the spray drying process of Clostridium butyricum was also investigated,and water content was found to be a vital factor in determining the storage stability of Clostridium butyricum dry powder.Maltodextrin as the spray drying carrier gave less water content thus a higher spore residual rate of the Clostridium butyricum dry powder compared with that of cornstarch,which demonstrates that maltodextrin is more suitable spraying drying carrier for Clostridium butyricum.
