| Orange head chinese cabbage(Brassica camperstris L.ssp.pekinesis(Lour.)Olsson)is named after the head heart for it is exposed to the air and gradually turns orange red,Orange head chinese cabbage is rich in nutritive value and gorgeous in color.that which is the research direction of Chinese cabbage quality breeding,The cytoplasmic male sterile line(CMS)does not require artificial detasseling,therefore it is easy to reproduce,saving labor costs,accelerating the speed of breeding,and more importantly,guaranteeing the high purity of the F1 hybrids In this study,The female parent is Green stem vegetables which sterility expresses stable,leaf avoids yellowing in seedlings stage under low temperature,Sterility strain rate and the fertility degrees were 100%.The excellent orange head chinese cabbage ‘420’ and ‘026’ as recurrent parent,By hybridization and continuous backcrossing,the cytoplasmic male sterile lines EruA420 and EruA026 of Chinese cabbage with genetic stability were obtained by combining field selection and molecular marker assisted selection.In order to evaluate the cytoplasmic male sterile line comprehensively,the flower organ observation,fertility investigation,and anther cytology observation were performed on the cytoplasmic male sterile lines EruA420 and EruA026 of the Chinese cabbage organg cabbage,and the time and mode of abortion of the anthers were established.At the same time,the cytoplasmic male sterility lines EruA420 and EruA026 were used as female parents and 18 elite inbred lines were used as male parents to prepare 36 hybrid combinations.Heterosis analysis and combining ability analysis were carried out to select excellent parents and cross combinations.The main results were as follows:1.In this study,EruA was used as the infertility source,and The orange head cabbage 420 and 026 were the recurrent parents.Through one cross and four backcrosses,combined with field phenotypic selection and molecular marker-assisted selection,the orange head cabbage cytoplasm was obtained male sterile lines EruA420 and EruA026.2.Observe the floral organs and investigate the fertility of the cytoplasmic sterile lines EruA420 and EruA026.The results showed that the stamens of the cytoplasmic male sterile lines EruA420 and EruA026 were severely aborted,and the anthers were triangular,white and pollen-free.The sterility and sterility rates reached 100%,and the stigma was exposed.Anther abortion of Chinese cabbage cytoplasmic male sterile line is the abortive occurs in the early stage of mononuclear,the tapetum expands and squeezes microspores in a radial direction,and finally the microspores and tapetums form a deeper staining material in the drug chamber.Unable to open,complete abortion.3.Analysis of heterosis of 36 hybrid combinations by using cytoplasmic male sterility lines EruA420 and EruA026 in organg cabbage cytoplasmic and crossed with 18 excellent inbred lines,found that all combinations exhibited significantly positive average parental advantage except for the number of outer leaves.Analysis Heterosis(HPH)showed that the combination of EruA026×AJQ49,EruA420×AJQ41,EruA420×RI8513,EruA026×RI223,EruA420×RI223 had over-parents heterosis in plant height and plant weight,and small heterosis in relatively plant width.Combination EruA026×AJQ49,EruA420×AJQ41 has a over-parent advantage in bulb height and bulb width.4.Combining effect analysis showed that for the female sterility line EruA420,negative effects were observed in plant height,plant weight,plant width,number of outer leaves,and head height,indicating that The sterile line EruA420 could be used as an alternative parents to choose the plants are short,compact variety.On the other hand,the male sterile line EruA026 has more positive effects of general combining ability and less negative effects for Chinese cabbage horticultural traits,and can be used as a breed with large size and high yield.For the parents RI8513,JB-1,and RI85,the general combining ability of the traits of weight and weight was greater,and it could be used as a breeding high-yield product.This experiment is also selected a better combination of yield traits EruA420×RI8513,EruA026×JHX-1,EruA026×JHX-3,EruA026×AJQ49. |
