| During pregnancy establishment,endometrial cells produce prostaglandins(PGs),including prostaglandin F2α(PGF2α)and prostaglandin E2(PGE2),which are critical for the establishment of pregnancy in ruminants.Both PGF2αand PGE2 are regulated by IFN-tau that is secreted by the trophoblast cells of ruminant animals,but the underlying mechanism remains unclear.C-Jun binding protein 1(JAB1)is a multifunctional binding protein involved in many biological responses and has an important influence on the growth and development of early embryos.Unfolded protein response(UPR)also plays an important role in mammalian reproductive physiology.In addition,studies have reported that JAB1 is also associated with UPR.The aim of this study was to investigate whether JAB1 and UPR play a role in IFN-tau regulation of PGs secretion in goat ESCs.The experimental results are as follows:(1)IFN-tau treated ESCs,ELISA detection of cell PGs secretion and found that IFN-tau reduced PGF2αsecretion and increased PGE2/PGF2αratio.PCR and Western Blot showed that IFN-tau can increase the expression of JAB1 and UPR makers.(2)We constructed goat JAB1 lentivirus interference vector.PCR and Western Blot detected interference efficiency.Results showed that the interference efficiency reached 80%.JAB1 interference significantly reduced UPR-related protein expression.(3)Under the condition of IFN-tau treatment of ESCs,ELISA assay confirmed that the interference of JAB1 resulted in the down-regulation of PGE2 secretion,and the ratio of PGE2/PGF2αsignificantly reduced.PCR and Western Blot showed that the expression of UPR-related proteins in JAB1 interference group significantly reduced.Pretreatment of ESCs with Tg activted UPR signaling pathway,the ratio of PGE2/PGF2αin the JAB1 interference groups increased.In summary,this study constructed a goat JAB1 lentivirus interfering vector and studied the functional roles of JAB1 and UPR in IFN-tau regulating PGs secreted by ESCs.It demonstrated that JAB1 and UPR mediate the secretion of PGs by ESCs.It expands our understanding of the mechanism of IFN-tau regulating PGs secreted by ESCs,and provides new insights into the biological functions of JAB1 and UPR. |
