Analysis On Quality Traits And Molecular Detection The Quality Related Genes And The Fusarium Head Blight Resistance Gene Fhb1 Of 246 Wheat Varieties

The wheat is one of important alimentray crop,good quality,high disease resistance of the germplasm resource is the material guarantee of the wheat breeding.246 wheat varieties from four province of Anhui,Jiangsu,Henan and Shanxi were analysed quality-related traits with Near infrared spectroscopy analysis,molecular marker-assisted selection and field survey.The results are as follow:1)In order to analysis the wheat quality and screen for high quality varieties for Huaibei area of Jiangsu province,246 wheat varieties from Huang-Huai region were analyzed by near-infrared spectroscopy technology.The results showed that the coefficients of variation were larger in development time,sedimentation value and stability time,which of volume weight,flour yield and water absorption were smaller.There were significant differences among Shanxi,Henan,Jiangsu and Anhui provinces in quality properties of wheat.Through screening of indicators in all varieties,40 varieties were suitable for strong gluten wheat,in which,17 from Henan,11 from Jiangsu,10 from Shaanxi and 2 from Anhui.2)The polyphenol oxidasw(PPO)gene related PP029?PP016?and PPO18 molecular markers were tested in 246 wheat varieties.The results showed with low activity Ppo-A 1b/Ppo-D1a accounted for 17.9%in wheat varieties of Zhengmai96177,Zhengmai111,Zhengnong 16,Zhengmai9694,Xinmai 13,Xinmai 19,Xinmai31,Yuzhanl,Luo8503,Zhongyu885 and Zhumai4 from Henan province;of Xumai541,Baomai2,Xumai6124,Xumai6052,Xumai7227,Xumai6951,Xumai7048,Xumai7086,Xumai8066,Xumai27,Huaimai11,Huaimai32,Huaimai33,Baofeng10-82,etc.from Jiangsu province of Xinong335,Xinongl 56 and soon from Shanxin provine,of Annong1208,etc.from Anhuiprovince.These varieties can effectively improve the color and luster of flour quality,will not cause a Browning phenomenon of the flour,also provides the wheat quality breeding germplasm resources.3)The predecessors involved molecular markers of Ax2,Dx5 and By8 genes were tested in 246 wheat varieties.There were no wheat variety with high molecular weight glutenin subunit(HMW-GS)wtih Ax2,Dx5 and By8;there were 30 wheat varieties with macromolecular protein gene wtih Dx5 and By8,the proportion of 12.2%;such as Wanmai 38,Huacheng3366,Xinong622,Shanyou225,Shannong534,Huaihel4131,Huaimai1558,Ruihua1588,Xumai 88,Huaihe14087,Ruihua0394,NingS-113,Baofeng 1289,Huaimai0458,Huaimai39,Huaimai 32,Huaimai 21,Huiamai18,Xu10-170,Xumai 7086,Bonong6,Guomai10,Yumai34,Luomai24,Zhouheimai1,Yumai29,Zhengyumai-9987,Xumai7086,Zhengnongl6,Zhengmailll,and Zhongmai 415 were contains the gene of Ax2 and By8.using molecular selection technology to obtain breeding material with high protein gene and to cultivate high quality wheat varieties.The methods can be effecttively used in the detection of macro-molecular protein gene in wheat.4)Molecular detection was performed on 246 wheat varieties using the molecular marker of Fhb1,a major gene of wheat against scab.The results showed that Fhb1 gene can efficiently improve the Fusarium Head Blight(FHB)resistance of Zhengmai96177,Xinmai23,Zhongmai875,Zhongmai415,Xumai6124 and Huaimai17 wheat cultivars from the Yellow-Huai Rivers valley winter Wheat zone.5)The results showed that Zhengmai96177 were wheat cultivar with strong gluten,high white flour and resistance FHB;Zhengnongl6 were wheat cultivars with strong gluten and high white flour,contains the gene of By8 and Dx5;Zhongmai415 were contains the gene of Ax2 and By8 with resistance FHB of wheat variety.
