In order to understand the genetic resources of the Wandong cattle,the information on the molecular genetics of the Wandong cattle was improved,providing reliable data for the Wandong cattle.Based on the methods of molecular biology,this study selected 56 healthy adult Dongdong cattle,measured their body measurements and body weight indexes,collected blood,and studied the genetic diversity of Wandong cattle.The study consists of the following three parts:Experiment ?:Genetic diversity of D-loop region and Cytb gene in Wandong cattleThe bovine ear tissues were collected,and the primers were designed to expand the D-loop region and Cytb gene of Wandong cattle.The genetic diversity of Wandong cattle was analyzed by sequencing.Results:The average content of A+T in the D-loop region was 61.4%.A total of 69 SNPs were detected.The N-J phylogenetic tree indicated that the Wandong Cattle were divided into two groups:the Bos taurus and the Bos indicus.There were 16 kinds of haplotypes accounted for 40.8%in the Bos taurus.10 kinds haplotypes in the Bos indicus account for 59.2%of the total,and the proportion of the ruminal line was high.The haplotype diversity(Hd)in the D-loop region was 0.824±0.00308 and the nucleotide diversity(Pi)was 0.02583±0.00184.The average A+T content of Cytb gene was 58.0%,showing A+T bias.A total of 8 SNPs were detected with 6 haplotypes,and the haplotype diversity(Hd)was 0.565±0.056.The diversity(Pi)is 0.00521±0.00046.The Cytb gene was divided into two lines:the Bos taurus and the Bos indicus,the Bos taurus accounted for 36.7%,and the Bos indicus accounted for 63.3%of the total.Experiment ?:Association analysis between the polymorphisms of GHR gene and MYH8 gene and growth traitsBody weight and body measurements were measured to analyze associations between GHR gene and MYH gene polymorphisms and body measurements.Results:There were polymorphisms in GHR gene and MYH8 gene,and there were three genotypes,all in Hardy-Weinberg equilibrium.In the Hind? cleavage site of GHR gene,the chest depth,girth circumference,and tendon length of AA individuals were significantly greater than those of AG and GG individuals(P<0.05).The height and height of AA individuals were greater than those of GG individuals(P<0.05)The body length of the GG type was smaller than that of the AA type and the GG type(P<0.05).At the mutation site of MYH8 gene,the body length of CC individuals was significantly smaller than that of CT and TT individuals(P<0.05).Experiment ?:Association analysis between UCP3 gene polymorphisms and serum biochemical markersBlood serum was collected and biochemical indicators were measured using biochemical kits.Correlation analysis between UCP3 gene polymorphisms and serum biochemical indicators was analyzed.Resullts:There were polymorphisms in the exon2 and exon3 mutations of UCP3 gene in Wandong cattle.Three genotypes were detected and all were in Hardy-Weinberg equilibrium.The mutation site of exon2 of UCP3 gene is in low polymorphism.The Bg? cleavage site of UCP3 exon3 is in a moderate polymorphism(0.25<PIC<0.5).There was a correlation between UCP3 gene and serum biochemical indicators:AA-type aminotransferase and aspartate aminotransferase in UCP3 exon3 were significantly higher than those in AG type and GG type(P<0.05).In summary,there are strong A+T bias in Wandong cattle,and there were two main types of groups:the Bos taurus and the Bos indicus.The genetic diversity of the Bos indicus is relatively scarce,but the Wandong cattle are more affected by the Bos indicus.There are three genotypes in the GHR gene and MYH8 gene mutations in Wandong cattle,all of which are in Hardy-Weinberg equilibrium and are all in moderate polymorphism.They can be used as molecular markers for breeding.There were polymorphisms in the exon2 and exon3 mutations of UCP3 gene in Wandong cattle,all of them were in Hardy-Weinberg equilibrium,and the population genetic balance was.The mutation site of UCP3exon2 is in a low polymorphism.The Bg? cleavage site of UCP3exon3 is moderately polymorphic(0.25<PIC<0.5).This site has the potential for selective breeding and can be used as a molecular marker.There is a correlation between UCP3 gene and serum biochemical indicators.UCP3 gene may regulate lipid metabolism and affect liver function.
