In order to study the effects of different concentrations of boron on the proliferation,apoptosis and immune function of spleen lymphocytes in rats and the application prospect of boron in porcine breeding.This test is to choose 2 months clean SD male rats,sterile separation of spleen lymphocytes,first of all determined by MTT and CCK-8 method for testing boron poisonous to the cells of lymphocyte function,FASC to detect lymphocyte proliferation and apoptosis of preliminary screening in vitro lymphocyte when the suitable dosage of boron,were 0.4 mmol/L and 40 mmol/L.And specificity of blocking estrogen receptor and MAPK signal pathway,and handle with boron,using FASC and Western blot,Quantitative Real-time PCR and ELASE detection proliferation,apoptosis,and the indicators of immune function about lymphocyte.At the same time,12 bogs of Duroc with similar weight were selected,and randomly divided into 3 groups.Each group of 4 head,Drinking water supplements 5 mg/L and 10 mg/L of boric acid for 4 weeks,the control group based diet feeding,at the end of the study,the vena cava blood was collected,to analyze the effect of boron on the physiological,biochemical and humoral immune functions of pig blood,the results are as follows:1,The toxic effects of boron on spleen lymphocytes in rats and the influence of the proliferation and apoptosisAdd 0.4 mmol/L of boric acid on lymphocyte proliferation,apoptosis and beneficial impact on the immune function,characterized by increased proliferation rate,apoptosis rate is reduced,gene and protein expression of PCNA and Bax quantity also increased significantly;When the addition amount reached 40 mmol/L,the toxic effect on lymphocytes was significantly increased,and the apoptosis rate of lymphocytes was increased.Meanwhile,the expression of caspase-3,Bax and P53 also increased significantly.2,Boron on the immune function of spleen lymphocytes of ratsThe influence of adding 0.4 mmol/L boric acid can make the lymphocyte subsets CD3+,CD4+and CD8+ T cells and CD4/CD8 ratio increases,while promoting lymphocyte secretion of IL-2,IL-4,INF-y and IgG;The addition of 40 mmol/L would decrease the immune function of lymphocytes,and the ratio of CD3+,CD4+and CD8+lymphocytes and CD4/CD8 decreased,and the secretion of IL-2,IL-4,INF-? and IgG decreased.3,Before and after the estrogen receptor blocking boron on lymphocyte proliferation,apoptosis,and the influence of immune functionIn vitro culture of rat spleen lymphocytes,add 0.4 mmol/L boric acid can promote lymphocyte proliferation,improve the immune function of lymphocytes;In contrast,the addition of 40 mmol/L of boric acid inhibited the proliferation of lymphocytes,promoted apoptosis,and reduced the immune function of lymphocytes.And estrogen receptor blockers MPP,PHTPP and G-15 to decrease the rate of lymphocyte proliferation and immune function at the same time,it can change the boron on the function of lymphocyte,and especially the influence of PHTPP,boron can be mediated by estrogen receptor on spleen lymphocyte proliferation,apoptosis,and immune function,and possibly ERp play the role of the more obvious.4,Before and after the MAPK block on lymphocyte proliferation,apoptosis,and the influence of immune functionWill the rats spleen lymphocytes in vitro,different concentrations of boron,found that after processing 0.4 mmol/L boric acid can be beneficial effect on lymphocyte proliferation,and the 40 mmol/L boric acid inhibits lymphocyte proliferation.We are adding boron treatment at the same time,blocking the MAPK and found SB203580,SP60012,SCH772984 can reduce the proliferation rate between groups,apoptosis rate increased,and the effect of change of boron on lymphocytes,prompt boron through MAPK pathway on the immune function of lymphocyte proliferation,apoptosis and effect;The effect of SP600125 is most obvious,so it is concluded that jnk-mapk may be the pathway of boron affecting lymphocytes.5.Effects of boron on the physiological,biochemical and humoral immune functions of pigsIn boars added 5 mg/L in drinking water and 10 mg/L of boric acid,can significantly increase the number of boars blood leukocyte WBC,at the same time also can improve the content of TP,TG and IgA,and 10 mg/L of boric acid can significantly improve the pig blood MCV,MCH,MCHC and the content of PLT,above all that boron can to the pig's blood physiological,biochemical and humoral immune function to produce beneficial effects.To sum up,the in vitro culture of rat spleen lymphocytes,boric acid can add 0.4 tendency for L on lymphocyte proliferation,apoptosis,and beneficial impact on the immune function,and when the adding amount of 40 tendency/L,show the toxic effects of boron,lymphocyte proliferation rate decreased,significantly increased rate of apoptosis,immune function is restrained.Estrogen receptor blockers MPP,PHTPP and g-15 can change the effect of boron on lymphocytes,so that the proliferation and apoptosis of lymphocytes in the same concentration of boron treatment group will be different.Likewise,specific blocking MAPK signaling pathways,SB203580,SP600125 and SCH772984 has also changed the trend of boron affect lymphocyte proliferation and apoptosis,prompt boron can be mediated by estrogen receptor activation of MAPK signaling pathway and regulation of lymphocyte proliferation,apoptosis,and immune function.In addition,PHTPP and SP600125 have great influence on lymphocytes,and we conclude that ER and JNK play a more important role in it.In boars added 5 mg/L in drinking water and 10 mg/L of boric acid,can significantly increase the number of boars the WBC in the blood and the content of TP,TG and IgA can explain boron humoral immune function of pigs to produce beneficial effects.
