Wheat is one of world's three ma.jor grain crops,and called as the second grain crop only to corn in world's production.Low temperature can cause enormous losses to yield and restrict plants' geographical distribution and growing development.Wheatear's morphology is one of important agronomic characters which can affect wheat's production,so researches on panicle characters have important significance on increasing wheat yield.By far,researches on molecular biology about wheat's cold-resistance are relatively thorough but not comprehensive,which still need more complete and systematic studies.Moreover,there are fewer studies on molecular biology about wheat's panicle characters.This research made use of Illumina HiSeq high-throughput sequencing platforms to sequence the transcriptomes and analyzed the results by bioinformatics software.Specific results are as follows:1.The cDNA library was built with roots,stems and leaves of Huaimai 18 and Jinghe 1 with and without cold treatment to sequence RNA-seq.Clean Data of ten samples were 7.16 Gb.Q30 were detected to>87.05%.TopHat was used to make Clean Reads from all the samples reattach to the genome of Chinese Spring Wheat to compare these sequences.As a result,the reattachment rates were around 63.13%?72.77%.2.Mapped Reads were analyzed by Cufflinks software,we optimized 14298 genetic structure and discovered 12897 new genes.Compared with sequences in NR,Swiss Prot,GO,COG,KOG and KEGG databases,9521 genes got functional annotations.In Cuffquant and Cuffnorm components,FPKM was adopted to analyze gene expression amounts and compared up-regulated and down-regulated genes between groups respectively.3.Functional annotations of differentially-expressed genes were classified and analyzed in GO database in KEGG database.There were 5429 genetic variations in roots,4652 genetic variations in leaves,4147 genetic variations in stems of Huaimai 18 and Jinghe 1 with cold treatment.The roots,stems and leaves compared between normal group and treatment group showed 5 genetic variations in Huaimai 18 and 74 genetic variations in Jinghe 1.Analysis about 6 genes of transcription factors or functions in two varieties of blades with q-PCR showed:Transcription factors were expressed highest in 4h,expression of functional genes increased relatively slowly,the expression trends between two samples were the same as the sequencing results.More than 20 genes about coldness response and adjustment were gotten in the classification of GO.Those genes need further analysis afterwards4.This research respectively measured and analyzed thousand seeds weight,grains per panicle,grain weight of wheat with oblong,clubs,elliptic and spindle panicle characters.Results showed that wheat with oblong panicle character was more suitable for high yield.Genes about flowers and embryonic development were conducted by hot spot clustering analysis and 70 new genes with annotations were discovered in GO database.
