Study On The Immunogenicity Of Combined Inactivated Mink Enteritis Virus And Canine Distemper Virus

Mink viral enteritis?MEV?and canine distemper?CD?are two acute and contagious diseases,which are important diseases of mink breeding industry currently.As yet,there are no effective treatments on them.Vaccination is the only preventive method.In this study,an immune experiment of the combined inactivated mink enteritis virus?MEV-L?and canine distemper virus?CDV-L?was performed in rat.After inactivation of MEV-L with 4:1000 formaldehyde under 37?for 48h and CDV-L with 1:1000 formaldehyde under 4?for 24h,the mixtures of MEV-L?HA 2⁶?and CDV-L(TCID₅₀10^-5/mL)were made in ratio of 1:1???,1:2???and 1:3???and10%aluminum hydroxide adjuvant was added to each mixture.Subsequently,three groups of rats were immunized with?,?and?mixture,respectively.The serum was prepared from blood of all rats at time points of 0,7,14,30,60 and 90 day and their titers of anti-CDV and anti-MEV were detected by serum neutralization test,hemagglutination inhibition test and indirect ELISA test.Results showed that HI titer against MEV at 7th day was up to 1:768???and the lowest was 1:171?III?.Indirect ELISA antibody titer reached 1:512???and the lowest was 1:128?III?.At 30th day the HI titers reached the highest level 1:15009???,indirect ELISA antibody titer reached 1:16384???.The titer of HI at 90th day when it was maintained at 1:2218???,indirect ELISA antibody titer remained at 1:2048???.The SN antibody titer of the CDV at7th day was more than 1:100???,lowest level of 1:37???.The highest titer of indirect ELISA antibody reached 1:256???,the lowest was 1:64???.SN antibody titer of the CDV at 30th day reached the highest level 1:2457???,indirect ELISA antibody titer reached 1:4096???.SN antibody titer at 90th day when CDV could be still maintained at 1:776???,indirect ELISA antibody titer remained at 1:1024 in group I.In conclusion,group I was the best in ratio of CDV and MEV.
