Chinese tallow tree(Sapium sebiferum(L.)Roxb.ndl),native to China,is rich in oil content and widely uesd in heavy industry,food industry and skin care products.Because of its beautiful tree shape and showy leave in the autumn,Chinese tallow tree is one of the highly ornamental colorful-leaf trees.However,due to the long history of cultivation and breeding of this specie,the classification and genetic relationship remain unclear.Research about genetic diversity of Sapium was still extremely limited.Here,we sequenced the specie to explore a batch of SSR primers with Illumina HiSeq2500 platform.Small sample was uesd to selected the SSR primers and genetic evaluation was given by means of these selected SSR primers.The conclusion as follow:1.Tender leaves were sequenced with Illumina HiSeq2500 platform,after assemblyed with trinity,59441 unigene were identified max as 6929 bp,min as 300 bp and 692 bp as average length.The function of 42020 unigene(70.69%)were annotated after blasted with Nr database from NCBI.31277 unigene were annotated into three parts after the Gene ontology analysis.2.In total,7344 of SSR loci were obtained using the MISA Software from the obtained unigene with occurrence 12.36%.With Software Primer 3,120 SSR primers were random designed based on the7344 SSR loci.10 SSR primer(8.33%)with polymorphism were selected using 6 eggplant materials.3.Thirteen populations in total 236 individual were testd with 10 pairs of polymorphism SSR marker for genetic evaluation.The result shows the germplasm exhibited the high level of variability,its coefficient Ne(1.9884)and Shannon Wiener index I(0.7045)had a similar symptom to other tree species like Liriodendron chinense(Hemsl.)Sarg.And germplasms dendrogram was given based on genetic similarity coefficient.The result proved thatthe classification and genetic relationship is disordered because of the long cultivation history.Here we firstly explore the EST-SSR of Sapium sebiferum(L.)Roxb.ndl,Which was valuable for the research of genetic diversity of Sapium,and the transcriptome data of Sapium can be referenced for the generic characters study.
