Lily is a world famous cut flower with rich fragrance,it has high ornamental value and economic value.In order to analyze the causes of lily fragrance and non-fragrance,the volatile constituents of 41 lily varieties from 12 hybrids were determined during full blooming period,the characteristics of fragrance constituents of lilies from different hybrids were studied,and the fragrance types of lilies were classified.The relationship between gene expression and fragrance release was found by real-time quantitative PCR analysis,combined with the differences of volatile species and release amounts of monoterpene synthase gene(TPS).We cloned promoter sequences of monoterpene synthase genes from two different aroma lily cultivars,and analyzed the sequence by bioinformatics elements to preliminarily study the promoter function of monoterpene synthase genes.The main results are as follows:1.46 aroma components were identified from 41 lilies of 12 hybrids.Terpenoids and benzenoids were the main groups.The main volatile components of Lilium are eucalyptol,(E)-?-Ocimene,linalool and methyl benzoate.The quantity and content of volatile components of non-scented Asiatic hybrids and Asiatic×Trumpet hybrids are significantly lower than those of other lilies.The hybrid lines obtained by cross-breeding again combine the aroma characteristics of their parents.2.According to the types and contents of main flower fragrance components and human olfactory senses,41 kinds of lilies were divided into six fragrance types: unscented type,cool type,fruit scent type,musky scent type,fruit-honey scent type and lily scent type.3.Real-time quantitative PCR showed that the expression level of monoterpene synthase gene in non-scented lily was significantly lower than that in scented.4.Three promoters of Lh TPS gene were cloned from non-scented AT6 and scented O6.The promoter sequences were analyzed by bioinformatics method.It was found that besides TATABOX and CAATBOX,there were many cis-acting elements related to light response,stress and hormone response.
