Traditional vegetable drugs has been used for centuries for the treatment of various diseases.However,the traditional way of treatment has decreased with the progress of science and technology,and at the same time the exploration and detection of herbal has increased.In the past few decades,it was more and more popular to extracted active ingredient from the plant,which would be used in the future.The polysaccharide is a kind of natural active substance which the biggest advantage is that the side effect is small and the source is wide,and it has many functions such as anti-virus,anti-oxidant,enhancing immunity and so on.The effects of the polysaccharide were greatly improved after sulfated modification.Ferula sinkiangensis K.M.Shen and M.nigra.Linn.were elected as the research object and polysaccharides were extracted to compare their abilities of anti-NDV and Immune enhancement.The polysaccharide with better effect was further purified and sulfated to compare the antiviral effect of modified polysaccharides.The experiment was divided into 5 parts:1.Total Ferula sinkiangensis K.M.Shen polysaccharide?cFSPt?and three different solvent extraction cFSP30,cFSP50,cFSP70;total M.nigra.Linn.polysaccharide?cMPt?,and cMP30,cMP50,cMP70 of were extracted using the methods of hot water extraction and ethanol precipition in this study.The highest content of polysaccharide was cFSP50,55.04%.All of the polysaccharides have typical characteristic absorption peaks of polysaccharides.2.In order to study the anti-Newcastle disease virus?NDV?and immune-enhancing function of cFSP in vitro,cell viability was determined by MTT,ntiviral and immune-enhancing effect of cFSP in vitro showed that,cFSPt and three different solvent extraction have anti-NDV and immune-enhancing activity.Among them,cFSP50 had the strongest anti-NDV activity,cFSPt had the strongest proliferation effect to spleen lymphocytes and peripheral blood lymphocytes.3.In order to study the anti-Newcastle disease virus?NDV?and immune-enhancing function of cMP in vitro,cell viability was determined by MTT.Antiviral and immune-enhancing effect of cMP in vitro showed that,cMPt and three different solvent extraction have anti-NDV and immune-enhancing activity.Among them,cMP70 had the strongest anti-NDV activity,MP30 synergistic PHA group had a certain effect on promoting lymphocyte proliferation,other groups of cMP had no obvious effect on lymphocyte proliferation.4.In order to get the modified product of Xinjiang asafetida polysaccharide and mulberry polysaccharide,first through D900,S-8 macroporous resin and Sephadex G-100 on cFSP50 cMP70,decolorization and purification.By the orthogonal experiment analysis,reaction temperature,reaction time and ratio of chlorosulfonic acid-pyridine polysaccharide DS,FSP50 effect the sugar content of MP70,were set 4 optimal modification conditions,sulfation,a total of 4 kinds of sulfated polysaccharides?sFSP0.7,sFSP1.9,sMP2.5,sMP1.3?.5.In order to study whether the modification can improve the activity of MP70 and FSP50,cell viability was determined by MTT.Antiviral and immune-enhancing effect FSP50,MP70 and 4 kinds of sulfated polysaccharides in vitro showed that,sMP2.5.5 possessed better antiviral activity,inhibition rate was87.69%.FSP50,MP70 and 4 kinds of sulfated polysaccharides were no obvious effect on the proliferation of peripheral blood lymphocytes.
