| 【Objective】To study the effects of micro-storage soybean straw on growth performance,blood index and antioxidant function of sheep,and the effects of VD3 on antioxidant function and apoptosis of sheep.【Method】The experiment was divided into two parts.RAMS weigh between 20 and 25 kg and 2months old were selected for 10 days of pre-feeding period and 70 days of experimental period.In the first experiment,10 sheep were randomly divided into 2 groups with 5 sheep in each group.Control group:basal diets+ air-dry soybean straw,experimental group: basal diets+ micro-storage soybean straw.In the second experiment,25 sheep were randomly divided into 5 groups with 5 sheep in each group.The amount of vitamin D3 added in the control group was 0 IU/kg,while the amount of VD3 added in the experimental group was 300,600,900 and 1200 IU/kg.Cell apoptosis was evaluated by TUNEL and Fluorescence Quantitative PCR assay and antioxidant capacity were detected by colorimetry.【Results】The results of the first experiment showed that the ADFI of the experimental group was significantly higher than the control group(P<0.05);The blood index of experimental group such as WBC,LY,GRAN,HGB,LY%,GRAN% and MID were significantly(P<0.05/P<0.01)higher than the control group.CAT activity of serum,thymus,liver,kidney and epididymis in the experimental group was significantly(P<0.05/P<0.01)increased.T-SOD activity of spleen,thymus,mesenteric lymph,testis and epididymis was significantly(P<0.05/P<0.01)increased.The GSH-PX activity of serum,liver,thymus,mesenteric lymph,testis and epididymis was significantly(P<0.05/P<0.01)increased.MDA contents of serum,spleen,testis and epididymis were significantly decreased(P<0.01).The results of the second experiment showed that WBC indexes of sheep in the test group with the addition level of VD3 at 300,600 and 900 IU/kg was significantly(P < 0.05)higher than that in the control group.LY index of sheep in the test group with the addition level of VD3 at 300 and 900 IU/kg was significantly(P<0.05/P<0.01)higher than that in the control group.The GRAN indexes of sheep in the experimental group with the addition level of VD3 at 600 and 900 IU/kg was significantly(P<0.05)higherthan that in the control group.The HGB indexes of sheep in the test group with the addition level of VD3 at300 and 600 IU/kg was significantly(P<0.01)higher than that in the control group.CAT activity(testes and epididymis),T-SOD activity(serum and liver),GSH-PX activity(spleen,testis and epididymis)were significantly(P<0.05/P<0.01)higher in each test group than the control group,MDA content(serum,liver,spleen,thymic,mesentery and epididymis)was significantly(P<0.05/P<0.01)lower than control group.VD3 was added 300 IU/kg,the CAT activity(mesenteric lymphatic),T-SOD activity(kidney,mesenteric lymphoid,testis and epididymis),and GSH-PX activity(serum kidney and mesenteric lymphoid)were significantly(P<0.05/P<0.01)higher than control group.VD3 was added at 600IU/kg,the CAT activity(serum),the T-SOD activity(epididymal)and the GSH-PX activity(liver)were significantly(P<0.05)higher than control group,and the MDA content(kidney)was significantly(P<0.05)lower than that of the control group.VD3 was added at 900 IU/kg,the CAT activity(serum,spleen,thymus),the T-SOD activity(kidney,mesenteric lymphatic)were significantly(P<0.05)higher than control group,and the MDA content(kidney)was significantly(P<0.05)lower than that of the control group.VD3 was added 1200 IU/kg,the CAT activity(serum,spleen,thymus),the T-SOD activity(kidney),the GSH-PX activity(liver)were significantly(P<0.05)higher than control group.VD3 added level of 300 IU/kg,the relative expression of BAX(epididymis),Caspase3(liver,kidney,testis),Caspase9(kidney,testis),VDR(testis)were significantly(P<0.05)lower than control group.VD3 added level of 600 IU/kg,the relative expression of BAX(thymus,epididymis),Caspase3,Caspase9 and VDR(kidney,thymus),Bcl2(testis,epididymis)were significantly(P<0.05)higher than control group,and the MOD value of liver,thymus and testis were significantly(P<0.05)than control group.VD3 added level of 900 IU/kg,the relative expression of VDR(thymus)was significantly(P<0.05)higher than control group,and the MOD of liver and kidney values were significantly(P<0.05)higher than control group.VD3 added level of 1200 IU/kg,the relative expression of VDR(epididymis)and Bcl2(spleen,testis,epididymis)were significantly(P<0.05)higher than control group,and MOD values of epididymis was significantly(P<0.05)than control group.VD3 added level of 600,900,1200 IU/kg,the relative expression levels of BAX,Caspase3 and Caspase9(liver,spleen,mesenteric lymphatics,testis),VDR(spleen,mesenteric lymphatics),Caspase3 and Caspase9(epididymis),BAX(kidney,mesenteric lymphatics)were significantly(P<0.01/P<0.05)higher than those in the control group,and the MOD values of spleen and mesenteric lymphatics were significantly(P<0.05)higher than control group.The relative expression of Bcl2 in sheepkidney of each experimental group was significantly(P<0.05)lower than that of the control group.【Conclusion】1.The feeding effect of micro-storage soybean straw was better than of dry soybean straw,the micro-storage soybean straw could improve the production performance,blood routine index and antioxidant capacity of sheep.2.Vitamin D3 could improve the blood indexes and antioxidant function of sheep,activate apoptosis genes,and induce morphological changes of cells,leading to apoptosis.The addition of 300 IU/kg Vitamin D3 could significantly improve the antioxidant capacity of sheep and reduce the apoptosis of tissue cells. |
