In order to clarify the genetic differences between the PFSP cultivars,the morphological markers and SSR markers were used to analyze the genetic diversity of the 75 PFSP cultivars released in China and 1 PFSP cultivar from Japan,and the quality traits of the 76 cultivars were also analyzed in this study.The main results are as follows:1.According to the guideline for the conduct of tests for distinctness,uniformity and stability— Sweetpotato(NT/T2429-2013),18 upper-ground traits and 3 underground traits of the tested cultivars were investigated.The results of PCA showed that the eigenvalues of the seven main components of the tested cultivars were greater than 1,and the cumulative variance contribution rate was 67.03%,the first,fourth and sixth principal components were related to the accumulation of anthocyanins;According to the NJ cluster analysis based on the Euclidean genetic distance,the 76 PFSP cultivars can be divided into 3 clusters at a genetic distance of 0.4789.And some geographically similar cultivars have a tendency to cluster together.2.The quality traits of crude starch,soluble sugar,reducing sugar and crude protein of the tested PFSP cultivars were determined by near-infrared analyzer.The results showed that there were significant differences among different cultivars.The composition of anthocyanin of the tested PFSP cultivars were determined by high performance liquid chromatography(HPLC),a total of 28 anthocyanin components were detected and Cynidin and Peonidin were the main components.3.Among 339 SSR primer pairs,124 primer pairs were screened with highly polymorphic.A total of 750 bands were amplified by using 124 primer pairs from 76 PFSP cultivars,among them,90.13% bands were polymorphic,and 2 to 18 bands were detected for each primer pair,with an average of 6.05.The Nei's genetic distance was calculated by NTSYS2.10 e,and that of 76 PFSP cultivars was between 0.0442 and 0.6010,with an average of 0.2921.The genetic distance among the PFSP cultivars from different sweetpotato growing areas(SPGA)was not obvious,but they were significantly larger between Ayamurasaki and the cultivars of domestic SPGAs;Using MEGA7.0.26 software and analyzing by NJ clustering method,the 76 PFSP cultivars can be divided into IX clusters at a Nei's genetic distance of 0.2253,and the IX cluster can be divided into 4 sub-clusters at a genetic distance of 0.2137,the cultivars with similar origins or the same parents had a tendency to gather together;Using STRUCTURE 2.3.4 software to analyze the population structure,it is found that the 76 cultivars were divided into 3 stable groups through analyzing the K value of the population structure.The reults indicated that there were 3 sources of variation in the genetic components of the tested materials.4.Two primer pairs(XZ-SSR15 and XZ-SSR63)with highly polymorphic were screened from 124 primer pairs,and DNA fingerprinting of the 76 PFSP cultivars based on the two primer pairs was constructed by capillary electrophoresis.The 76 cultivars can be completely distinguished by using the two primer pairs.
