Avian influenza is caused by the avian influenza virus,and result in sickness a class of acute exposure to avian infectious sexually transmitted diseases,and lead to a huge loss of poultry industry in China,but also caused a serious threat for the safety of our people's.At present,the HA and NA subtypes avian influenza virus were 18 and 11,of which H5,H7 subtype of avian influenza virus the strongest pathogenicity,but also the current focus of prevention and treatment.The wild waterfowl is a natural host of bird flu,greatly boosting the spread of avian influenza virus.In order to reduce the impact of avian flu on humans,countries around the world,we are highly concerned about the avian flu,through a variety of means to strengthen the monitoring of wild waterfowl,to take active measures to minimize the risk.1.Background Survey of Avian Influenza in Jiangsu ProvinceIn order to determine the wild water avian influenza virus infection in Jiangsu Province,from December 2014 to March 2017,in Yancheng red-crowned crane wetland ecological tourism zone,Dafeng Elk Reserve,Xuyi Hongze Lake,Xuzhou and other places more than 20 species of wild Waterfowl about 605 swabs were sampled.Detection of universal primers using avian influenza detection by E.Hoffmann et al.The results show that the samples collected are mainly Crane-shaped and wild-shaped water birds,involving large migratory birds and common wild waterfow;through the detection of wild birds in Jiangsu Province to show the avian influenza virus,the rate of virus was 8.26%;the virus carrying rate was 23.08%in Gallinula chloropus,and 10.94%and 10.71%,in Anas platyrhynchos,Cygnu respectively2.Quantitative Assessment of the Risk of Avian Influenza in Wild Water in Jiangsu ProvinceThe risk assessment model was established with reference to the animal disease risk analysis theory designated by the OIE and the modeling conditions of @Risk software.The assessment of the model includes:the distribution of avian influenza in China,the avian influenza infection,the impact of temperature on the spread of avian influenza,the spread of waterfowl migration,and the prevention and control of avian flu in five aspects.By running the @Risk software,the risk assessment results were obtained after 10,000 iterations.The results showed that the risk value of wild water avian influenza was 2.003 in Jiangsu,which was between the moderate risk and the high risk,and it was a high risk level.Among them,each risk factor,the regression coefficient of Jiangsu wild waterfowl is the highest in the whole risk assessment,the regression coefficient is 0.8,the infection rate of migratory migratory birds in Jiangsu,and the distribution of suitable poultry habitat in the spring and autumn season also contributes more to the risk assessment.The results of the assessment remind us that we should strengthen the surveillance of wild waterfowl in Jiangsu Province and formulate preventive measures and countermeasures for avian flu.3 Genome Sequencing and Genetic Analysis of H3N8 SubtypeAvian Influenza Virus Isolated from MallardAn avian influenza virus was isolated from a mallard in Jiangsu wetland.To investigate the origin,molecular evolution and epidemic characteristics of the strain,the entire of the isolate was subjected to genome sequencing with genetic evolution analysis.Wild waterfowl fecal samples detected by A common type of avian influenza fluorescent RT-PCR reagents.The positive samples were inoculated with the 10-day-old SPF chicken embryos;Collect allantoic fluid within 24-96 hours;Hemagglutination and the sequence was determined to be analyzed.The results showed that the virus belongs to the H3N8 subtype of avian influenza virus,named:A/Mallard/Xuyi/14/2015(H3N8).It indicated that the amino acid motif of the HA gene cleavage site was PEKQTR ? GLF,which is the typical characterization of the low pathogenic avian influenza virus.Glycosylation sites of HA and NA did not have mutations;NA resistant amino acid sites appeared mutation site;The 627 and 701 amino of PB2 protein is glutamate(Glu)and aspartic acid(Asn),which are AIV signature amino acid.In GenBank,The strain sequence alignment analysis showed that:10 strains with the highest affinity for each gene located in the China Eastern migratory area.The strains are low pathogenic avian influenza virusthat gene is derived from different regions of the viral genes which mainly from China Eastern migratory zone Jiangsu province located in the eastern zone of migratory birds,rich wetland resources,a wide variety of birds,and we must further strengthen bird flu prevention and immunization by monitoring migratory birds.
