Abscisic acid(ABA)is an important plant endogenous hormone,and it is also a plant-derived biochemical pesticide.It plays an important role in plant growth and response to external abiotic stress and biotic stress.Brown planthopper,Nilipavata lugens St?l(BPH)is a piercing-sucking pest that ingests phloem sap of rice.Previous studies had found that exogenous ABA increased the area of rice callose deposition and increased resistance to BPH.Studies indicated that one of the mechanisms of ABA defense pathogens is the formation of callose,and the other is participating in the biosynthesis of jasmonic acid(JA),which activates the expression of JA-mediated resistance genes.BPH is different from chewing pests,and the sucking behaviour may be similar to pathogen infection.ABA may interact with JA in rice resistance to BPH.In this paper,the usage of osaba8ox3 T-DNA insertion mutant further studied rice resistance mechanism to BPH.Metabolomics analysis in rice after exogenous ABA and JA treatment,BPH feeding were used to study the interaction between ABA and JA.The experimental results were as follows:1.Study of the resistance of osaba8ox3 mutant to BPHOsABA8ox3 is a key gene in ABA hydrolase which plays an important role in controlling rice ABA content and drought stress,but there is no research on resistance to insects.The phenotypes of oscaba8ox3 mutant and Zhonghua 11 wild type(WT)were significantly different,the expression of ayaoa8ox3 mutant was significantly higher than that of WT,and the expression of hydrolase gene was significantly lower than that of WT.Electrical penetration graphs(EPG)showed the duration of sucking phloem(N4 wave)of osaba8ox3 mutant was significantly lower than that of WT,indicating that the resistance of osaba8ox3 mutant to BPH was significantly increased.Without BPH feeding,the callose deposition of osaba8ox3 mutant was significantly higher than that of WT,and after BPH feeding,the osabca8ox3 mutant and WT callose deposition area were significantly higher than the control,which showed that OsABA80x3 played an important role in rice resistance to BPH.2.Comparative study of metabolomics analysis in rice after hormone treatment and BPH feedingThe contents of sugar and amino acids in rice decreased significantly after ABA and JA treatments,while the contents of sugars and terpenes in rice after BPH feeding increased and the contents of organic acids decreased significantly.There are important metabolic pathways in rice.JA treatment and BPH feeding influenced the metabolism of GABA bypass in rice,ABA and JA treatments influenced rice ABC transporter pathway,and ABA treatment and BPH feeding affected pentose phosphate pathway.It can be seen that ABA not only indirectly participates in the pathway of JA' resistance to BPH,but also directly participates in rice resistance to BPH.
